Protara's January 2025 presentation says cash runway reaches into 2027 when it counts $102.7 million of gross proceeds from a recent offering. Its balance sheet figure is $81.5 million at September 30, 2024, and its bladder cancer efficacy data come from a small single-arm trial.

The runway claim

Protara Therapeutics, Inc. (TARA) said in a corporate presentation dated January 13, 2025, furnished in an 8-K, that its cash runway reaches into 2027. That figure includes $102.7 million of gross proceeds from what the company calls a recent public offering. The presentation lists $81.5 million of cash, cash equivalents and investments as of September 30, 2024, which does not yet include those proceeds.

The offering added 14.1 million common shares and 2.3 million pre-funded warrants. Before it, the company had 30.3 million common share equivalents on an as-converted basis at September 30, 2024, made up of 20.6 million common shares, 8.0 million from preferred stock and 1.7 million pre-funded warrants. The total after the offering is 46.7 million.

The presentation excludes 10.8 million common warrants issued in an April 2024 private placement. Those warrants are exercisable at $5.25 a share and expire at the earlier of April 10, 2027 or 90 days after the company announces a six-month complete response rate of at least 42 percent among at least 25 BCG-unresponsive patients in its ADVANCED-2 trial.

The presentation names no underwriter, placement agent or counsel for the offering and gives none of its terms beyond the share and warrant counts.

The bladder cancer data

The lead program, TARA-002, is an inactivated strain of Streptococcus pyogenes that the company calls a broad immunopotentiator. It is being tested in non-muscle invasive bladder cancer. The efficacy figures come from the ADVANCED-2 trial, with data cut off on November 19, 2024.

Of the 24 patients enrolled, 20 were evaluated for high-grade complete response at month three and later, 18 at month six and three at month nine. The company reports a combined 72 percent six-month response and a 70 percent response at any time across BCG exposures. The 100 percent durability from three to six months rests on 9 of 9 patients, and the 80 percent reinduction salvage rate rests on 4 of 5.

These are interim results from a single-arm study with no control group. The presentation describes a registrational design and a planned futility analysis, but it does not show that the results would hold in a larger trial.

The safety data

The company says it has seen no grade 2 or greater treatment-related adverse events to date. The presentation's own table is less clean. It shows 3 of 24 subjects with grade 3 adverse events of any kind, 7 with grade 2 and 16 with events of any grade. Related events occurred in 3 subjects, all at grade 1. A footnote lists two urinary tract infections and one urosepsis as serious events and marks them as not drug related.

The presentation calls the safety profile favorable. That is the company's characterization of an interim dataset of 24 patients, which is too small to establish the rate of adverse events in a larger population.

The other programs

The presentation also covers intravenous choline chloride for patients dependent on parenteral support, for which it plans a pivotal study called THRIVE-3 with 120 patients. It describes a Phase 2 trial of TARA-002 in pediatric lymphatic malformations, listed as a single-arm, open-label study of 29 patients. Both are early or still in planning, and their results are not in the presentation.

What the runway depends on

The runway depends on the $102.7 million gross figure. The presentation does not give the net proceeds after fees, the current burn rate or the operating loss, and it does not explain how cash will be split across three programs with separate trials. The registrational trial in BCG-unresponsive bladder cancer has no approval timeline in the presentation.

What comes next

The presentation lists a futility analysis, more ADVANCED-2 interim data, a combination trial in BCG-naive patients and the THRIVE-3 pivotal study as milestones, several described only as planned or expected. Exercise of the April 2024 warrants at $5.25 each would add shares.
