Ascendis is profitable, but one drug carries most of the revenue
Ascendis is profitable, with three marketed TransCon drugs and a clear European decision due this quarter. Its risks are reliance on YORVIPATH for most revenue, a royalty to BioMarin and the end of the Novo partnership in obesity.

Opinion only. The Investor Rank is aibio.space's opinion of the company's business position, drawn from the public sources on this page. It is not a recommendation to buy, sell or hold any security and it is not investment advice. It does not consider the share price, valuation or your circumstances, and it can change at any time. Payments from companies never affect it. How the rank works.
- Technology
- Proven platform. TransCon underpins three marketed products, YORVIPATH, SKYTROFA and YUVIWEL, and a combination of two of them is heading to phase 3.
- Odds
- Favorable near term. The EU decision on YUVIWEL is due in Q4 2026 after a positive CHMP opinion, and rare disease filings historically win approval 93.6% of the time.
- Balance sheet
- Solid. Cash was €812 million at June 30 with positive operating cash flow, and the convertible notes were converted into equity.
- Main risk
- Concentration. YORVIPATH is about 80% of product revenue, and the BioMarin royalty and Novo rights reversion add cost and strategic questions.
- Watch next
- European Commission decision on YUVIWEL. Ascendis expects it in Q4 2026, with a German launch in Q1 2027 if approved.

Ascendis and BioMarin agree binding term sheet to settle all litigation
Ascendis said in a 6-K that on August 31, 2026 it announced a binding term sheet with BioMarin for a global settlement and license covering YUVIWEL and navepegritide-related products. BioMarin grants a non-exclusive worldwide royalty-bearing license and agrees to dismiss all proceedings, and Ascendis agrees to dismiss its proceedings against BioMarin. Ascendis would pay royalties of 20% on U.S. net sales and 18% in the European Union, South Korea and Brazil through May 20, 2030.
- Litigation
BioMarin ITC patent complaint against Ascendis reached trial in April 2026
In its interim report furnished August 13, 2026, Ascendis said BioMarin filed a complaint with the U.S. International Trade Commission alleging infringement of U.S. Reissue Patent No. 48,267, and that trial took place in April 2026. Ascendis started a related action in the U.S. District Court for the Northern District of California, which is stayed pending the ITC proceedings. Ascendis says it believes BioMarin's claims lack merit. The August 31 term sheet later addressed these disputes.

UPC dismisses BioMarin patent case against Ascendis after EPO revocation
Ascendis reported in its 20-F that the EPO Technical Boards of Appeal revoked BioMarin's EP3175863 patent in its entirety on October 16, 2025. Ascendis also instituted proceedings in the Danish Maritime and Commercial High Court over divisional applications of the revoked patent.

Fact-check-o-meter
Ascendis messaging mostly matches its filings, but the combo data and the profit headline need context
Ascendis's releases are largely in line with its interim report and 20-F, and the company is profitable with no going-concern language. The gaps are in emphasis: a one-time voucher sale drives the headline operating margin, recent releases drop the word 'accelerated' from YUVIWEL's U.S. approval, and the 'tripling of efficacy' claim for the combination rests on a 21-child open-label trial with no concurrent control. On Aug. 31, 2026 Ascendis filed a 6-K saying it had entered into a binding term sheet to settle all litigation with BioMarin, including the U.S. ITC case tried in April 2026, with a definitive agreement still to come.

Q2 operating margin includes a one-time voucher sale
The company said: "Operating profit for the second quarter of 2026 totaled €220 million, reflecting a margin of 65%. On a non-IFRS basis, operating profit was €92 million*, reflecting a margin of 27%*." source
The record says: The interim report says the PRV sale closed in the second quarter and "The income from sale of the PRV, net of transaction costs was € 158.1 million and is presented as other operating income". It is also "included within cash flows from operating activities in the statements of cash flows". source
Our view: The release does show the non-IFRS figure next to the IFRS one, so nothing is hidden. Still, the 65% margin and the first-half operating cash flow both carry a voucher sale that cannot repeat, and the 27% non-IFRS margin is the better guide to the run rate.

Recent releases drop 'accelerated' from YUVIWEL's U.S. approval
The company said: "Making YUVIWEL available in select International Markets countries through early access programs using U.S. FDA approval." source
The record says: The February approval report states the drug was approved "under the FDA's Accelerated Approval Program" and that "Continued approval for this indication, which was based on an improvement in annualized growth velocity, may be contingent upon verification and description of clinical benefit in confirmatory trial(s)." The interim report repeats: "In February 2026, we received accelerated approval of YUVIWEL". source
Our view: The August results release and the August 6 program update describe YUVIWEL's FDA status without the word accelerated. The approval rests on growth velocity, a surrogate, so U.S. revenue depends on confirmatory data the later releases do not discuss.

'Tripling of efficacy' comes from an open-label, single-arm phase 2
The company said: "sustained increases in ACH-specific height Z-score, indicating a tripling of efficacy compared to TransCon CNP monotherapy." source
The record says: The release itself calls COACH "an ongoing prospective Phase 2 open-label trial" with cohorts of "N=12" and "N=9". The ClinicalTrials.gov record lists a single-group design with no masking, an actual enrollment of 22, and a primary endpoint of growth velocity at week 52 measured as "cm per year, compared to navepegritide alone". source
Our view: With no concurrent control arm, the comparison to monotherapy is against other trials, and the treatment-experienced cohort is small. The registry's enrollment of 22 also differs from the 21 children in the release. The planned phase 3 is where the 'tripling' claim gets a real test.

BioMarin patent litigation, including the U.S. ITC case, is covered by an Aug. 31 binding term sheet to settle
The record says: The interim report says BioMarin filed "a complaint filed with the U.S. International Trade Commission ("ITC") related to alleged infringement of U.S. Reissue Patent No. 48,267. Trial in the ITC took place this April 2026." Ascendis's own suit in the U.S. Northern District of California "has been stayed in view of the pending ITC proceedings." The European '863 patent was revoked on October 16, 2025 and the UPC case dismissed. A 6-K filed on Aug. 31, 2026, after the interim report, says Ascendis "entered into a binding term sheet for a global settlement and license agreement with BioMarin" and that the term sheet "provides the terms and conditions upon which the companies agree to resolve all litigation and disputes between the companies". Ascendis would pay royalties of 20% in the United States and 18% in the European Union, South Korea and Brazil on net sales of navepegritide-related products through May 20, 2030, and each side would dismiss its proceedings against the other. The 6-K refers to an anticipated definitive agreement. Our view: The August results release and the October CHMP release mention litigation only in generic forward-looking language and do not mention the term sheet. The interim report says Ascendis believes BioMarin's claims lack merit, while the later 6-K describes a settlement with royalty payments, so the definitive agreement and its final terms are the facts to follow.
