Summit's FDA date arrives before its global lung data

Summit is a one-drug company facing its first FDA decision on Nov 14 with a survival result that has improved but has not been formally significant. The larger question is whether the global first-line trials replicate the China data. Until then the company funds itself by selling stock, and its latest 10-Q says that is not yet enough for twelve months.

Technology
Strong in China, unproven in the West. Ivonescimab beat tislelizumab on survival in Akeso's HARMONi-6 trial (hazard ratio 0.66), but global data are mixed.
Odds
Hard to call. The HARMONi survival hazard ratio is 0.76 without a disclosed confidence interval, and the FDA reportedly asked for significant survival.
Balance sheet
Funded by stock sales. $690.7 million at June 30, a $380 million ATM program, and going concern language in the 10-Q.
Main risk
Western lung data. The HARMONi-3 squamous interim PFS missed its higher bar, and the final analysis is due in the second half of 2026.

Some gaps

Summit's survival story reads stronger in releases than in its own 10-Q

Summit's releases describe the HARMONi survival data as consistent and confidence building ahead of the Nov 14, 2026 FDA date. The 10-Q adds that the FDA said a statistically significant survival benefit is necessary in this setting, which the primary analysis did not reach, and the later figures carry only nominal p values. The going concern language in the 10-Q was absent from the earnings release, though the $2 billion AstraZeneca investment announced in September changes the cash picture.

  1. Press release vs filing · weight high

FDA's survival bar is in the 10-Q, not the release

The company said: "these data reinforce our confidence in the HARMONi results as we work toward the potential approval of ivonescimab in the U.S." source

The record says: The 10-Q states: "The FDA noted that a statistically significant OS benefit is necessary to support marketing authorization in this setting and the PFS results from this study may not be sufficient to support marketing authorization." It also says the primary OS analysis showed a positive trend "without achieving a statistically significant benefit with a hazard ratio of 0.79" (p=0.057). source

Our view: The Sept. 15 release names the Nov 14 PDUFA date but does not repeat the FDA's stated requirement. That requirement is the main test the application faces, and the release's upbeat framing reads differently once it is known.

  2. Science · weight high

Later survival looks carry nominal p values, and the western subgroup still crosses 1

The company said: "With longer follow-up, western patients replicated the survival improvement seen in Asian patients, further reinforcing the regional consistency of the efficacy results observed in the global HARMONi study." source

The record says: The same release's table gives the June 2026 global hazard ratio as 0.76 with "nominal p=0.0151" and the western subgroup hazard ratio as 0.76 with a 95% confidence interval of 0.52 to 1.10 (n=83 vs 82). Asian patient data were locked at April 2025. ClinicalTrials.gov lists OS in the ITT population as a primary outcome alongside PFS, with primary completion on April 12, 2025. source

Our view: The protocol's OS test was the April 2025 analysis, which missed. Extra data cuts with nominal p values are supportive at best, and a western subgroup interval that includes 1 is not the same as replication.

  3. Press release vs filing · weight medium

Earnings release led with cash; the 10-Q states going concern doubt in its notes, MD&A and risk factors

The company said: "Summit Closes Second Quarter with $690.7 Million in Cash and Investments" source

The record says: The 10-Q filed the same day says: "These conditions raise substantial doubt about our ability to continue as a going concern." On Sept. 28, 2026 Summit announced a $2.0 billion AstraZeneca investment in convertible preferred stock; the 8-K says closing was expected "within five business days" and conversion depends on HSR clearance and a charter amendment raising authorized shares. source

Our view: The July release did not mention the going concern language. The AstraZeneca money, if closed, should address it, and the next 10-Q will show whether the language is removed.

  4. Science · weight medium

Most clean survival wins come from China-only Akeso trials

The company said: "including recent positive results from HARMONi-2 and HARMONi-GI1 in biliary tract cancer" source

The record says: The release itself says HARMONi-2 "is a single-region, multi-center Phase III study conducted in China and sponsored by Akeso, with all relevant data exclusively generated, managed, and analyzed by Akeso" and that its OS result came from an interim analysis of "a secondary endpoint". The 10-Q says the global HARMONi-3 squamous interim PFS look faced "a meaningfully higher bar" and the iDMC "recommended that the study continue as planned". source

Our view: Summit discloses the China-only design plainly, which is fair. The global HARMONi-3 study has not yet reported a win, so the China results remain the main support for broader use.

  5. Legal and disclosure record · weight low

Derivative suit over insider notes was dismissed in May 2026

The record says: Legal Proceedings in the 10-Q: on March 17, 2025, Rainaldi Revocable Trust filed a derivative lawsuit in the Delaware Court of Chancery over the December 2022 notes from Mr. Duggan and Dr. Zanganeh. "On May 27, 2026, Plaintiff and the Defendants stipulated to voluntary dismissal of Plaintiff's claims with prejudice as to Plaintiff and without prejudice as to any other stockholder of the Company". No securities class action or enforcement action is disclosed. source

Our view: The one disclosed suit is closed as to that plaintiff. The 10-Q describes the dismissal as without prejudice as to any other stockholder.
