Xenon has an epilepsy filing and a depression problem
Xenon has strong epilepsy data, a filed NDA and about $1.2 billion in cash, which together carry the company well into a possible launch. The depression program is now the open question after neuropsychiatric events led to a pause. Epilepsy approval odds look good on the base rates, while the psychiatry upside is less clear.

Fact-check-o-meter
The epilepsy case is strong; the psychiatry safety message and trial size need more detail
Xenon's epilepsy numbers match across its releases, though 'highest ever' is a cross-trial claim. The September release says the new psychiatry adverse events are consistent with azetukalner's known profile and also says they were not seen in Phase 2, and it gives no counts. X-NOVA2 will now read out with about 360 patients against a registry target of 450.

Psychiatry pause: 'consistent with the known profile' and 'not previously seen' in one release
The company said: "The observed events, the rate of these events and their severity, are consistent with the known safety and tolerability profile of azetukalner and its mechanism" source
The record says: The same release continues: "however, such adverse events had not previously been seen in the Phase 2 X-NOVA clinical study in MDD." The Aug. 6 10-Q, filed six weeks earlier, said: "while adverse events in our X-TOLE and X-NOVA clinical studies were generally mild or moderate in severity, there can be no guarantee that we will observe a similar tolerability profile of azetukalner in our ongoing Phase 3 clinical studies". source
Our view: The 10-Q risk factor did warn that Phase 3 tolerability might differ, and the company paused enrollment with its data safety monitoring board. But the release calls the events neuropsychiatric without saying which ones, how many patients had them, or at what dose, so 'consistent with the known profile' cannot be tested from the public record.

X-NOVA2 will read out with about 360 of a planned 450 patients
The company said: "X-NOVA2 enrollment has reached approximately 80% of its initial target of 450 patients, which is sufficiently powered to demonstrate a clinically meaningful change in the study's primary endpoint of HAM-D17." source
The record says: The same release describes the program as "approximately 450 patients per study". ClinicalTrials.gov, updated Oct. 13, 2026, lists X-NOVA2 (NCT06775379) as active, not recruiting but still shows enrollment as 450 (estimated), double-masked, with primary outcome "Change from baseline in the Hamilton Depression Rating Scale, 17-item (HAMD-17) score at Week 6". source
Our view: Closing enrollment at about 80% of target is the company's decision, and the power claim is the company's own statement. Depression trials are noisy and placebo response is high, so a smaller trial is more exposed to a miss. The registry shows the study as no longer recruiting but has not updated the enrollment target.
Science · weight medium
'Highest placebo-adjusted efficacy ever' is a cross-trial claim
The company said: "to our knowledge, show the highest placebo-adjusted efficacy ever observed in a pivotal epilepsy study" source
The record says: The release compares X-TOLE2 (week 12, placebo-adjusted median change -42.7% at 25 mg) with Phase 2b X-TOLE, "which demonstrated a -34.6% placebo-adjusted MPC in the 25 mg dose group over 8 weeks". It describes participants as having "a median of five prior ASMs" and a baseline of 12.75 seizures per month. source
Our view: The result is large and the primary and key secondary endpoints were met. But 'ever' compares different trials, populations and trial lengths, with no head-to-head test. The company hedges with 'to our knowledge'; the comparison still carries more weight in the headline than it can bear.
Science · weight medium
'Generally well-tolerated' alongside 14.5% discontinuations at 25 mg
The company said: "azetukalner was generally well-tolerated with a consistent safety profile observed in both studies" source
The record says: The X-TOLE2 release reports dizziness in 20.5% of azetukalner patients versus 3.2% on placebo, and "14.5% of participants in the 25 mg group, 4.8% in the 15 mg group, and 3.2% in the placebo group had a TEAE leading to treatment discontinuation." source
Our view: Serious adverse events were low and similar across groups (5.6%, 3.2% and 2.4%), which supports the wording. The 25 mg dose, the one with the best efficacy number, also had the highest dropout from side effects. An FDA label will have to weigh both.
Press release vs filing · weight low
'Runway into 2029' appears in the release but not the 10-Q
The company said: "Based on current operating plans, Xenon anticipates having sufficient cash to fund operations into 2029." source
The record says: The 10-Q says the company "expects the cash, cash equivalents and marketable securities to be sufficient to fund current operations for at least the next 12 months from the issuance of the financial statements" and adds that "our estimates and assumptions may prove to be wrong". source
Our view: The two statements are not in conflict, since 12 months is the accounting test. The 2029 date was given on Aug. 6, before the Sept. 17 pause, and the Sept. 17 release does not restate it, and the cash is $1,245.1 million.
Legal and disclosure record · weight low
No securities litigation or enforcement disclosed in the latest filing
The record says: The Q2 10-Q, filed Aug. 6, 2026, says: "We are not presently a party to any legal proceedings that, in the opinion of our management, would reasonably be expected to have a material adverse effect on our business, financial condition, operating results or cash flows if determined adversely to us." source
Our view: That filing predates the Sept. 17 enrollment pause, so it cannot speak to anything since. We found no later filing that discloses a case, and the 10-Q's securities class action risk factor is generic.
