Beam's base editing works in the clinic; selling it is the next test
Beam has two programs near registration and clean biology in both. The bigger questions are commercial: whether risto-cel can win patients in a slow sickle cell market, and whether the debt-backed runway holds if milestones slip.

- Technology
- Strong early biology. Risto-cel lifted fetal hemoglobin above 60%, and BEAM-302 cut mutant Z-AAT 84% at 60 mg.
- Odds
- Good at filing, early elsewhere. Base rates put approval near 91% once a BLA is filed, but BEAM-301 and BEAM-304 are in phase 1/2.
- Balance sheet
- Funded with debt support. $1.15 billion at June 30, 2026, and runway into mid-2029 depends on more Sixth Street draws.
- Main risk
- Commercial uptake. Risto-cel would launch second, behind Casgevy, in a market slowed by chemotherapy conditioning.
- Watch next
- Risto-cel filing. Updated BEACON data and a possible BLA are due by year-end 2026.

Fact-check-o-meter
Beam's messaging mostly matches its filings, but the runway and the BEAM-302 safety story need the fine print
Beam's releases and its second-quarter 10-Q tell the same basic story, and the 10-Q discloses no material legal proceedings. The gaps are in framing: the mid-2029 runway counts $200 million of debt that only unlocks on risto-cel regulatory milestones, and the September BEAM-302 release left out the multi-dose cohort where the 10-Q reports a Grade 4 liver enzyme rise. The AATD data are open-label biomarker results, and the 10-Q and the September release report 60 mg AAT levels with different assays (LC-MS versus turbidimetry), so the two means are not directly comparable.

Runway leans on milestone-gated debt
The company said: "together with an additional $200 million expected to be drawn from the company's facility with Sixth Street, will fund anticipated operating expenses and capital expenditure requirements into mid-2029" source
The record says: The 10-Q says the facility offers "a potential additional $ 100 million draw upon the acceptance by the U.S. Food and Drug Administration, or FDA, of the Company's biologics license application, or BLA, submission for risto-cel prior to a certain date" and a second $100 million "upon the FDA's approval of the risto-cel BLA prior to a certain date". It reports an effective interest rate of 10.5% at June 30, 2026, and its own going-concern horizon is that cash "will be sufficient to fund its operations for at least the next 12 months". source
Our view: The release presents the $200 million as expected, while the 10-Q makes both tranches depend on FDA accepting and then approving risto-cel by dates it does not disclose. If the BLA slips, part of the mid-2029 runway goes with it, so the runway is better read as tied to the risto-cel timeline.

BEAM-302 safety summary leaves out the redosed patients
The company said: "Data from patients in the multi-dose cohort (n=3) and the Part A expansion cohort (n=6) were not included." source
The record says: The 10-Q, describing the February 10, 2026 data cut, states: "In the multi-dose cohort (n=3), following the second dose of BEAM-302, patients experienced Grade 2 IRRs, one patient had Grade 4 ALT and Grade 3 AST elevations, and one patient had a Grade 2 ALT elevation." It adds that the elevations were asymptomatic and did not require treatment. source
Our view: The September release says plainly that the multi-dose and Part A expansion cohorts were not included and describes single-dose BEAM-302 as well tolerated, which the 10-Q data also support. But a reader of the release alone would not learn that one of the three patients in the multi-dose cohort had a Grade 4 ALT elevation after the second dose, which matters if some patients ever need a second dose to reach target levels.

60 mg AAT figures in the 10-Q and the September release use different assays
The company said: "mean and median were 14.4 µM and 15.2 µM, respectively" source
The record says: The 10-Q reports the February 10, 2026 cut for the 60 mg Part A cohort (n=6): the steady-state circulating total AAT mean (LC-MS) "was 16.1 µM in the 60 mg cohort (follow-up ranging from 5-12 months)". Its footnotes define steady state as running from Day 28 to Month 12 (or the last visit if earlier) and say levels were measured with a liquid chromatography-mass spectrometry assay. The September release, for the June 24, 2026 cut, gives a mean of 14.4 µM, defines steady state as running from Day 28 to the patient's last visit, says "Circulating total AAT levels were measured using turbidimetry", and states "Data were consistent with topline results announced in March 2026." source
Our view: Both figures sit above the 11 µM protective threshold the company uses, and the company calls the data consistent with March 2026. Because the assay, the follow-up window and the data cut all differ, the 14.4 µM figure cannot be read as a decline from 16.1 µM. The release does not say how turbidimetry and LC-MS results compare, which matters for the 12-month AAT biomarker the registrational cohort will be judged on.

Registrational AATD cohort is open-label and judged on a biomarker
The company said: "The cohort is designed to support a potential accelerated approval path in the United States." source
The record says: ClinicalTrials.gov lists NCT06389877 as single group, allocation not applicable, masking none, with an estimated 192 participants and primary outcomes including "Mean total and functional AAT in blood" over 1 year. The 10-Q says the plan is "a primary endpoint of AAT biomarkers evaluated over 12 months" in about 50 patients in "a pivotal expansion of the ongoing open-label Phase 1/2 trial". source
Our view: An accelerated approval on blood AAT levels would rest on a surrogate, with no placebo arm and no lung function endpoint in the registrational cohort. That is a normal path for a rare disease, but it leaves the clinical lung benefit to be shown later.

Risto-cel data come from a small single-arm trial
The company said: "Ristoglogene autogetemcel (risto-cel, formerly known as BEAM-101) is an investigational autologous cell therapy with a potential best-in-class profile for the treatment of sickle cell disease (SCD)." source
The record says: ClinicalTrials.gov lists BEACON (NCT05456880) as single group with masking none. The 10-Q states: "one patient died four months after risto-cel infusion due to respiratory failure that was determined by the investigator to be likely related to busulfan conditioning and deemed unrelated to risto-cel." source
Our view: Best-in-class is the company's own description. BEACON is registered as a single group study, so any comparison with other sickle cell therapies is cross-trial. The 10-Q discloses a death that the investigator judged likely related to busulfan conditioning and unrelated to risto-cel.
Legal and disclosure record · weight low
No litigation or enforcement disclosed
The record says: Part II, Item 1 of the 10-Q for the quarter ended June 30, 2026: "We are not currently subject to any material legal proceedings." source
Our view: No securities litigation or enforcement disclosed in the latest filing. The risk factors flag possible patent challenges in base editing as a general risk, not as a pending case.
