Intellia's first approval looks likely; the open questions are sales and nex-z's liver signal

Intellia has clean phase 3 data for a one-time HAE therapy and an FDA decision due March 10, 2027, at a stage where most filings get approved. The harder questions are how fast lonvo-z sells against established chronic drugs and whether nex-z's liver signal can be managed with HLA screening.

Technology
Validated in phase 3. One dose of lonvo-z cut HAE attacks 87% against placebo, with no serious adverse events in the treated arm.
Odds
High for lonvo-z, open for nex-z. Filed applications have a 90.6% historical approval rate, while nex-z faces a phase 3 outcomes trial and an unresolved liver question.
Balance sheet
Funded into 2028. $628.4 million at June 30 plus a $75 million OrbiMed draw, with up to $225 million more tied mostly to lonvo-z milestones.
Main risk
Nex-z liver safety. An HLA allele linked to the worst enzyme rises could narrow eligible patients or prompt new regulatory demands.
Watch next
PDUFA on March 10, 2027. Before that, MAGNITUDE-2 enrollment completion in the second half of 2026.

Some gaps

Lonvo-z messaging holds up; the nex-z safety story reads softer in the release than in the 10-Q

Intellia's HAE claims match its filings and the HAELO registry record, and the trial design is strong for a rare disease study. The August release framed the nex-z HLA finding as a safety insight without the clinical hold and patient death that the 10-Q recounts, and gave no numbers behind the finding. The one securities class action on file was dismissed in June 2026.

  1. Press release vs filing · weight medium

HLA finding presented without the hold and death context

The company said: "gained important new genomic insights that enhance our understanding of nex-z's safety profile" source

The record says: The 10-Q states the FDA placed a clinical hold on both phase 3 INDs on October 29, 2025 after "a report of Grade 4 liver transaminase elevations and increased total bilirubin in a patient who was dosed with nex-z in the MAGNITUDE trial. This patient passed away on November 5, 2025." It adds that the patient died of septic shock from a perforated duodenal ulcer, with a course that "also included acute liver injury", that the FDA lifted the clinical holds in the first quarter of 2026, that the FDA lifted the clinical holds in the first quarter of 2026, that the FDA lifted the clinical holds in the first quarter of 2026, that the FDA lifted the clinical holds in the first quarter of 2026, that the FDA lifted the clinical holds in the first quarter of 2026, and that the HLA work is ongoing "As we engage with the FDA regarding this finding". source

Our view: The release is accurate but reads as good news. The filing ties the HLA analysis to a serious liver event and a patient death, although it also says the FDA lifted both holds in the first quarter of 2026. The 10-Q describes the discussion with the FDA on the HLA finding as ongoing, so it does not show what screening, if any, will be required.

  2. Science · weight medium

HLA link is a retrospective genomic analysis with no numbers disclosed

The company said: "The analysis revealed that the highest observed liver transaminase elevations were in patients carrying one specific human leukocyte antigen (HLA) allele." source

The record says: The 10-Q describes "a comprehensive genomic analysis of more than 600 patient samples across nex-z clinical trials" and the same conclusion about "one specific human leukocyte antigen ('HLA') allele." Neither document names the allele, says how many patients carry it, or gives enzyme levels by genotype. source

Our view: A look-back analysis can point to a cause but cannot prove that screening out carriers removes the risk. Until Intellia shows carrier frequency and event rates by genotype, the size of the problem in a 1,200 patient ATTR-CM trial is hard to judge.

  3. Science · weight low

HAELO design is strong; durability data are still short

The company said: "HAELO met its primary endpoint with an 87% reduction (p<0.0001) in mean monthly attacks" source

The record says: ClinicalTrials.gov lists HAELO as randomized and quadruple masked with 80 participants (actual), primary outcome "Time-normalized number of Investigator-confirmed HAE attacks" over "From Week 5 through Week 28", matching the release. Long-term outcomes run "From Week 5 through Week 104" and study completion is estimated for 2027-09. The 10-Q says lonvo-z is designed to drive "potentially lifelong reduction in kallikrein levels". source

Our view: The headline number rests on a blinded, placebo-controlled trial with a primary endpoint that matches the registry, which is better evidence than most rare disease filings. The open question is durability: the controlled window is 24 weeks, while the pitch is a one-time, lifelong treatment.

  4. Press release vs filing · weight low

Runway claim matches the filing, with conditions and later debt

The company said: "The company's existing cash resources are expected to fund its operations at least into 2028" source

The record says: The 10-Q bases the runway on June 30 cash plus "the net proceeds from our April 2026 equity offering" and collaboration funding, excluding milestones and "any potential commercial revenues from lonvo-z", and adds "We have based this estimate on current assumptions that may prove to be wrong". A September 4, 2026 8-K added an OrbiMed credit facility with a covenant to "maintain liquidity of at least $50 million in controlled accounts until the FDA approves the BLA for lonvo-z". source

Our view: The release and 10-Q agree; there is no going-concern language. The new secured loan adds covenants and events of default that the August runway statement predates, so the balance sheet now carries obligations that equity alone did not.

  5. Legal and disclosure record · weight low

Class action dismissed; derivative and patent suits pending

The record says: Legal Proceedings in the Q2 2026 10-Q lists three matters. Gonzalez v. Intellia Therapeutics, Inc., No. 1:25-cv-01353 (D. Mass.), a putative securities class action filed February 11, 2025 over statements about NTLA-3001; the court "fully granted the defendants' motion to dismiss on June 18, 2026, which decision the plaintiffs did not appeal." Aiello v. Bhanji et al. (C.A. No. 2025-0543-BWD), a stockholder derivative suit filed May 15, 2025 in the Delaware Court of Chancery over non-employee director pay, in discovery. BlueAllele Corp. v. Intellia Therapeutics, Inc., a patent suit filed July 8, 2024 in the U.S. District Court for the District of Delaware, in discovery. source

Our view: The securities case is over. The remaining suits concern director compensation and the company's bi-directional insertion template technology, and BlueAllele seeks damages and an injunction. None was mentioned in the releases reviewed.
