Intellia's first approval looks likely; the open questions are sales and nex-z's liver signal
Lonvo-z goes to the FDA with strong phase 3 data and a March 2027 date. The value of the company now rests on launch uptake and on what regulators make of an HLA allele tied to nex-z liver risk.
The technology
Intellia edits genes inside the body. Its two late-stage drugs are one-time infusions that use CRISPR/Cas9 to switch off one gene for good. Lonvo-z targets KLKB1 to permanently lower kallikrein in hereditary angioedema. Nex-z targets TTR to stop production of the protein that builds up in transthyretin amyloidosis, and is partnered with Regeneron.
The HAE data are strong. In the phase 3 HAELO trial, one 50 mg dose cut monthly attacks by 87% against placebo, and 62% of treated patients were free of attacks and other therapy for six months, against 11% on placebo. Moderate and severe attacks fell 91%. No serious adverse events were seen in the lonvo-z arm.
The odds
Lonvo-z is under priority review with a March 10, 2027 action date and no advisory committee planned for now. In the BIO, Informa and QLS data set for 2011 to 2020, 90.6% of filed applications won approval, and 93.6% in rare diseases. A clean phase 3 with no serious adverse events in the treated arm supports that base rate. The new parts are regulatory: this would be the first in vivo CRISPR approval, so manufacturing and long-term follow-up terms have no precedent.
Nex-z is a phase 3 bet with a safety question attached. The same data set puts the odds of moving from phase 3 to a filing at 57.8% and 52.4% from phase 3 to approval. The FDA held both trials from October 2025 after a Grade 4 liver event and lifted the holds in the first quarter of 2026. In August, Intellia linked the worst liver enzyme rises to one HLA allele and is discussing it with regulators.
The money
Cash, equivalents and marketable securities were $628.4 million on June 30, 2026, after about $195 million net from an April stock sale. The company guides that this lasts at least into 2028, before any lonvo-z revenue. The second-quarter net loss was $106.6 million.
In September Intellia added a credit facility of up to $400 million from OrbiMed. It drew $75 million at closing. Another $75 million comes with FDA approval of lonvo-z and $120 million more with revenue targets. The loan carries interest of at least 9.15% a year, is secured by substantially all assets including IP, and requires $50 million of liquidity until approval.
The risks
Nex-z safety is the largest. The HLA finding could explain the liver signal, or regulators could ask for more. Screening out carriers would shrink the market; Zacks reported the allele in about 12% of samples.
Lonvo-z faces a market with recent entrants. GEN listed Andembry, Ekterly and Dawnzera among 2025 approvals. A one-time edit is a different ask for patients whose disease is already controlled, and 20% of HAELO enrollees reported full control on prior prophylaxis.
The debt adds pressure after approval, when covenants switch to revenue or market capitalization tests. The company also defends a patent suit, BlueAllele v. Intellia, filed in July 2024 in Delaware, where the court denied Intellia's motion to dismiss.
Our read
Intellia reported the first phase 3 data for an in vivo gene editing therapy and has a near-term FDA date with high historical odds. The company has also staffed up for launch, adding a new chief medical officer who starts October 26. The value question has moved from whether CRISPR in the body works to how fast a one-time HAE therapy sells against good chronic drugs, and whether nex-z can clear its liver question in a 1,200-patient outcomes trial. The balance sheet covers the launch window on the company's own guidance, and the OrbiMed tranches are built to grow only if lonvo-z does. Execution risk now sits mostly in commercial uptake and in the HLA talks with regulators.
