Ascendis is profitable, but one drug carries most of the revenue

YORVIPATH sold €252 million in the second quarter. A European decision on YUVIWEL is due in the fourth quarter.

Ascendis Pharma has moved from a research company to a profitable one in about a year. It now has three marketed TransCon medicines and, as of September, no big-pharma partner in obesity. The open questions are whether its rare disease franchise can keep growing past one dominant product, and what it does with the metabolic rights it has taken back.

The technology

TransCon is the company's platform for turning drugs into longer-acting forms. Its three products are YORVIPATH (palopegteriparatide, for hypoparathyroidism), SKYTROFA (lonapegsomatropin, a growth hormone) and YUVIWEL (navepegritide, for achondroplasia). The EMA describes navepegritide as a prodrug that releases CNP, and the CHMP recommended it on October 15 after a trial showing 1.49 cm per year more growth than placebo.

The next step is combinations. In the phase 2 COACH trial, 21 children given TransCon CNP plus TransCon hGH kept growth velocity at or above the 97th percentile of average-stature children for 78 weeks. That is an open-label result without a control group.

The oldest product has the longest record. In June, Ascendis reported week 182 data from the PaTHway trial of YORVIPATH in adults with hypoparathyroidism: 86% of patients met a multi-part response endpoint, and all of them were independent of active vitamin D. Long follow-up of that kind matters because payers and doctors want proof that the benefit lasts.

The odds

Ascendis is past the highest-risk stage for its main assets. For rare disease, the BIO, Informa and QLS data put the move from phase III to filing at 60.4% and approval after filing at 93.6%. Overall, 17.0% of rare disease programs that start phase I reach approval.

The nearer events are regulatory and commercial. The European Commission decision on YUVIWEL is due in the fourth quarter. A phase 3 trial of the CNP and hGH combination is expected to start enrolling in the same quarter, and a hypochondroplasia trial in the second half of the year. The company also finished enrolling the infant trial, reACHin, which supports planned filings.

The money

Ascendis reported €812 million in cash on June 30, up from €616 million at the end of 2025. Second-quarter product revenue was €315 million and first-half operating cash flow was €274 million. The company converted $575 million of convertible notes into equity in May, removing that debt, and on September 14 it authorized a buyback of up to $400 million.

Quarterly profit was helped by a €158 million priority review voucher sale, so the IFRS figures run ahead of the underlying business. The non-IFRS operating profit was €92 million, a 27% margin.

The risks

Concentration is the first. YORVIPATH was €252 million of the quarter's €315 million product revenue, about 80%. Second, the company agreed to pay BioMarin royalties of 20% in the US and 18% in other markets on navepegritide until May 2030, under a term sheet that still needs a definitive agreement. Third, the loss of the Novo partnership means any obesity program is now Ascendis's to fund or partner again. YUVIWEL sales are still small, at €8 million in the second quarter, and the US approval is an accelerated one.

Our read

The company is financially sound and has proven it can launch rare disease drugs. The commercial record with YORVIPATH is strong, and YUVIWEL is on its way in Europe. The weak points are dependence on one product and the open question of strategy in obesity and cardiometabolic disease, where Ascendis now has rights but no partner. The Commission decision and early YUVIWEL sales will show how much of the growth can come from a second product.
