MetaVia completes dose titration in Phase 1 Part 3 study of DA-1726

Tickers MTVA

All active patients in both Part 3 cohorts of the DA-1726 obesity study reached target doses of 48 mg and 64 mg, and topline data remains on track for the fourth quarter of 2026.

MetaVia Inc. (Nasdaq: MTVA) said on July 9, 2026 that all enrolled active patients in Part 3 of its Phase 1 clinical trial of DA-1726 for the treatment of obesity had completed dose titration and were receiving their highest target doses in both study cohorts. The release says topline data remains on track for the fourth quarter of 2026.

The Part 3 design

Part 3 consists of two 16-week titration cohorts designed to evaluate one-step and two-step dose escalation strategies. The strategies aim to reach higher target doses safely while further optimizing tolerability. In Part 3A, patients titrated from 16 mg to 48 mg. In Part 3B, patients titrated from 16 mg to 32 mg and then to 64 mg. Part 3A uses 16 mg for 4 weeks followed by 48 mg for 12 weeks. Part 3B uses 16 mg for 4 weeks, 32 mg for 4 weeks and 64 mg for 8 weeks.

The trial plans approximately 40 obese, otherwise healthy adult subjects across the two parts, with approximately 20 subjects per part, randomized 4:1, with 16 active and 4 placebo. The study will assess safety, tolerability, pharmacokinetics and pharmacodynamics of DA-1726. Primary endpoints include adverse events, serious adverse events, treatment-emergent adverse events and adverse events leading to treatment discontinuation. Secondary and exploratory endpoints include pharmacokinetic profiling and measures of weight, waist circumference, body mass index and other cardiometabolic health. The release refers readers to ClinicalTrials.gov under identifier NCT06252220.

The earlier data

The company cited its previously reported Phase 1 multiple ascending dose study. That study included 9.1% mean weight loss at the 48 mg dose after 8 weeks of treatment, meaningful reductions in waist circumference, improved glycemic measures and early signs of direct liver benefit.

Hyung Heon Kim, President and Chief Executive Officer of MetaVia, said in the release that "The successful completion of dose titration across both Part 3 cohorts represents an important milestone for the DA-1726 development program." He said the company believes its titration strategy could be a meaningful competitive advantage over currently marketed obesity therapies, a view the release does not support with comparative data.

About DA-1726

The release describes DA-1726 as a novel GLP1R and GCGR dual agonist for the treatment of obesity and Metabolic Dysfunction-Associated Steatohepatitis (MASH), to be administered once weekly under the skin. In preclinical mouse models, the company says DA-1726 produced improved weight loss compared with semaglutide. The release says that in the Phase 1 multiple ascending dose trial in obesity, the 32 mg dose demonstrated best-in-class potential for weight loss, glucose control and waist circumference reduction.

The annual report, filed March 26, 2026, said the company planned to start Part 3a and Part 3b of the Phase 1 trial in April 2026, and that the data readout for both parts is planned for the fourth quarter of 2026.

On the financial side, the annual report shows research and development expense of $6.8 million for 2025, compared with $21.6 million for 2024, and a net loss of $13.0 million for 2025. Cash used in operations was $15.7 million for 2025, against $24.7 million for 2024.

What completing titration does not tell us

Finishing titration is a dosing step, not a result. The release reports no weight, safety or tolerability data from Part 3, and each cohort has about 16 active subjects and 4 on placebo, so the readout will rest on very small numbers. The annual report says the titration studies aim to remove moderate adverse effects seen at the 48 mg dose, which means tolerability at that dose is still the open question. The earlier 9.1% weight figure came from a different, non-titrated cohort. The sources available at the announcement date give no year-end 2025 cash balance; year-end 2024 cash and cash equivalents were $16.0 million, and 2025 cash used in operations was $15.7 million. Kim said the company remains focused on reporting topline data in the fourth quarter of 2026.
