Bolt Biotherapeutics, Inc. (Nasdaq: BOLT) reported financial results for the quarter ended March 31, 2026 on May 12, 2026, and provided a business update. Cash, cash equivalents and marketable securities were $23.9 million at March 31, 2026. The company said that balance is anticipated to fund operations into 2027, including completion of the dose escalation portion of its ongoing BDC-4182 Phase 1/2 study.
The platform and lead program
Bolt describes itself as a clinical-stage biopharmaceutical company developing novel immunotherapies for cancer. Its Boltbody ISAC platform pairs a tumor-targeting antibody with a non-cleavable linker and a proprietary immune stimulant. The immune stimulant is designed to recruit and activate myeloid cells, which release cytokines and chemokines that attract other immune cells.
The lead program, BDC-4182, is a next-generation ISAC that targets claudin 18.2, which the release calls a clinically validated target. The release says BDC-4182 demonstrated significant anti-tumor activity in preclinical models, including models with low claudin 18.2 expression. It also says the candidate induced immunological memory and outperformed cytotoxic ADCs in those models.
Bolt has implemented step-up dosing, which the company says has been used commercially for T-cell engagers, to reach higher doses safely. Patients with gastric and gastroesophageal cancer are still being enrolled. Chief Executive Officer Willie Quinn said the company is encouraged by what it has seen so far in patients treated with BDC-4182, and that the candidate has a unique mechanism of action.
Programs on hold
Two other ISAC programs are on hold, one targeting CEA and one targeting PD-L1. The release says both are close to clinical candidate selection, and that Bolt plans to resume them once BDC-4182 demonstrates proof of concept for the ISAC approach.
The CEA ISAC pairs a fully human antibody with high affinity and selectivity for CEACAM5 with a proprietary next-generation TLR7/8 agonist, linked through a non-cleavable linker. The release says it induced complete and durable anti-tumor responses in preclinical models, and that it was well tolerated in a non-GLP toxicology study.
First-quarter financial results
Collaboration revenue was $26,000 for the quarter, compared with $1.2 million in the same quarter of 2025. The release says the earlier revenue came from services performed under the R&D collaborations as the company fulfilled its performance obligations.
Research and development expenses were $4.8 million, down from $9.5 million. The release attributes the decrease to lower salary and related expenses from the restructuring, reduced clinical trial expenses and lower research and development spending overall. General and administrative expenses were $2.8 million, compared with $3.8 million, mainly because of lower salary and related costs after the restructuring.
Loss from operations was $7.6 million for the quarter, compared with $12.1 million a year earlier.
Full-year context
The 10-K filed March 12, 2026 reports 2025 revenue of $7.7 million and research and development expense of $28.5 million, compared with research and development expense of $57.5 million in 2024. The net loss was $33.4 million in 2025 and $63.1 million in 2024. Cash used in operations was $39.9 million in 2025 and $61.3 million in 2024.
What the release does not show
The release gives no clinical data. The CEO says he is encouraged by what he has seen, but no response rates, patient numbers or safety findings are reported. The efficacy claims, including that BDC-4182 outperformed cytotoxic ADCs, are preclinical, and the CEA ISAC's tolerability was tested only in a non-GLP toxicology study. Initial clinical data are expected in the third quarter of 2026,, and the study is still in dose escalation.
The balance sheet is tight. Cash of $23.9 million is anticipated to fund operations only into 2027, and the release ties that runway to completing dose escalation. Bolt used $39.9 million of cash in operations in 2025, and first-quarter collaboration revenue fell to $26,000 from $1.2 million. Spending fell mainly because of a restructuring. The CEA and PD-L1 programs stay on hold until BDC-4182 shows proof of concept, so the company's pipeline depends on one candidate.
