Aptevo Therapeutics Inc. said on September 24, 2026 that it is optimizing several trispecific candidates for reliable, scalable manufacturing, a step it describes as essential before future human testing. The company expects to enter Investigational New Drug (IND) enabling studies for its lead trispecific candidate, APVO451, in the first half of 2027. The release is an exhibit to an 8-K.
What the trispecific work involves
The release explains that each trispecific molecule can combine three complementary functions: recognizing cancer, activating an immune attack, and helping to overcome the defenses cancer uses to survive. Aptevo says its protein engineering and process development expertise allows it to bring these functions together in one stable molecule.
Peter Pavlik, PhD, Senior Director of Protein Engineering at Aptevo, said: "Trispecifics open an exciting new path in cancer drug development by allowing us to engineer several complementary functions into one therapy."
Aptevo says it is moving its trispecific candidates beyond molecular design by developing manufacturing methods that can produce them for human testing. According to the release, showing that these complex molecules can be manufactured consistently helps reduce a major development risk and prepares the lead candidate for the steps required before human testing.
The company also says the same expertise may support additional trispecific candidates and strengthen its ADAPTIR-FLEX platform. It expects to present more details on the ongoing development work at an upcoming international scientific conference. The release does not name that conference.
The company and its pipeline
Aptevo describes itself as a clinical-stage biotechnology company developing novel multispecific immunotherapies for cancer. The company's 10-K says it has two clinical candidates and six preclinical candidates, built on the ADAPTIR and ADAPTIR-FLEX platforms, which it wholly owns. The 10-K says the platforms are designed to generate monospecific and multispecific antibody candidates that enhance the immune system against cancer cells.
The 10-K also says preclinical candidates based on the ADAPTIR-FLEX platform are advancing in its pipeline. Its lead clinical candidate, mipletamig, is a CD123xCD3 T cell engager being evaluated in the RAINIER trial for frontline acute myelogenous leukemia with venetoclax and azacitidine. The 10-K says the RAINIER program was initiated in August 2024.
Clinical candidate ALG.APV-527 targets 4-1BB and 5T4 and is being evaluated for multiple solid tumor types. The 10-K says that, based on the safety and tolerability results from mipletamig, which uses a unique and proprietary CRIS-7 binding domain, the company has built out its CD3 engaging portfolio to five molecules.
The financial position
The XBRL data show research and development expense of $14.5 million for 2025, compared with $14.4 million for 2024. The net loss was $26.0 million in 2025 and $24.1 million in 2024. Operations used $25.6 million of cash in 2025 and $23.8 million in 2024.
Year-end 2025 cash and cash equivalents were $21.6 million, according to a 10-Q filed August 14, 2026, compared with $8.7 million at year-end 2024. The XBRL data also show $9.8 million of cash and cash equivalents at June 30, 2026.
What the release does not show
The release contains no data of any kind on APVO451: no animal results, no activity or safety findings, and no manufacturing yield. Its claims that manufacturing "helps reduce a major development risk" and that trispecifics are "gaining attention" are the company's framing. The step it announces is preclinical work that comes before an IND filing, and an IND is only permission to start a first human trial. The release gives no date for any human dosing, and the first-half 2027 target is the company's own expectation, not a commitment.
The company also has to pay for it. Cash used in operations was $25.6 million in 2025, while cash and cash equivalents were $9.8 million at June 30, 2026. Aptevo has no product revenue to cover that gap, so continued work depends on new financing that could dilute current shareholders. Its clinical candidates, mipletamig and ALG.APV-527, remain the company's only programs in people.
