Alaunos combines two ALN1003 mouse studies into one preclinical readout

Alaunos Therapeutics, Inc. (Nasdaq: TCRT) reported an integrated preclinical readout for ALN1003 on May 26, 2026. The release combines two non-Good Laboratory Practice (non-GLP) diet-induced obesity (DIO) mouse studies that the company had reported on March 2, 2026 and May 18, 2026, together with cross-study analyses in a non-confidential slide deck published the same day. The company says the data show treatment-associated changes in body weight and food intake, body composition, insulin resistance biology, adipose endocrine signaling and hepatic biology.

The two studies

The 48-day study, DIO Study 1, gave ALN1003 orally twice daily at a single dose level. Mean percent change in body weight for treated mice peaked at -12.9% compared with DIO controls. The release calls this the study in which the fullest expression of the multi-axis profile was observed.

The 18-day study, DIO Study 2, gave ALN1003 in drinking water at low, medium and high dose levels. The middle and high doses were 3 and 9 times the low dose. The release notes that reduced water consumption and apparent dose aversion may confound attribution of effects solely to drug exposure in this administration route. Alaunos says it is pursuing formulation optimization and dedicated pharmacokinetic and tolerability work.

Results and limits

In Study 2, fat as a percentage of body weight changed by -21.9% at Day 17 in the high-dose group, while lean tissue as a percentage of body weight rose 17.2%. Blood glucose in the high-dose group was 197 mg/dL, compared with 320 mg/dL in DIO controls.

In Study 1, the company reported lower fasting insulin and lower HOMA-IR after adjustment for body fat (ANCOVA p=0.0006). It also reported numerically lower leptin, significantly higher adiponectin and a significantly higher adiponectin-to-leptin ratio.

ALN1003 reduced liver weight by 43% in Study 1. In a blinded review of selected liver samples from both studies, mean NAS scores were 2.7 in the 48-day study and 1.3 in the 18-day study, compared with a score of 5 for controls. Alaunos says these limited pilot pathology findings do not establish MASLD resolution, fibrosis reversal, inflammation improvement or clinical efficacy.

Study 1 recorded mild, short-term, reversible hypolocomotion after approximately one-half of dose administrations, with no similar observations in control animals. In Study 2, two mice in the high-dose group were noted to be slightly dehydrated during the pharmacokinetic portion of the study.

Holger Weis, chief executive officer, said: "What we believe makes the ALN1003 preclinical profile distinctive is consistency of treatment-associated changes across several metabolic readouts." He added that the company's findings are early, non-GLP results in a mouse model.

Finances and what is still unproven

Alaunos reported a net loss of $4.2 million for 2025, compared with $4.7 million for 2024. Research and development expense was $1.4 million in 2025, up from $362,000 in 2024. The accumulated deficit stood at $924.6 million at the end of 2025.

The company plans additional preclinical studies and chemistry, manufacturing and controls activities to characterize ALN1003's pharmacology, evaluate exposure-response relationships and optimize formulation approaches. It has completed initial non-GLP, single-dose pharmacokinetic studies in large animals, and says the observed profile is consistent with further evaluation of a twice-daily dosing schedule. Small-scale manufacturing has started, and a larger production run may follow, subject to available capital, technical feasibility and development priorities. A computational chemistry program to design, synthesize and test ALN1003 analogs has also begun.

The release states that ALN1003 has not been evaluated in human clinical trials, and that its safety and efficacy in humans have not been established.

The readout joins two studies of different length, dose route and dosing, so the combined profile is not a single controlled result. The weight-loss figure comes from a single dose level in 48-day Study 1, while the drinking-water Study 2 carries the confound the company itself describes. The company calls the data early and non-GLP, and the p-values in the 10-K are nominal and unadjusted for multiple comparisons. The release gives no cash balance or runway, and the 2025 net loss and $924.6 million accumulated deficit show a company that will need new money to fund the work it plans.
