Foghorn Therapeutics Inc. is a clinical-stage precision therapeutics biotechnology company developing medicines that correct abnormal gene expression by selectively targeting the chromatin regulatory system. Its collaboration with Eli Lilly and Company dates from December 2021 and covers SMARCA2 and an undisclosed oncology target. Lilly has initiated a Phase 1 dose escalation trial of FHD-909 (LY4050784), a selective ATPase inhibitor of SMARCA2, and dosed the first patient in October 2024. The company says the trial is focused on SMARCA4-mutated cancers, with non-small cell lung cancer as the primary patient population. Foghorn's discovery programs also target CBP, EP300 and ARID1B, and the company says it has the potential to file four Investigational New Drug applications over the next two years.
