Ensysce Biosciences, Inc. is a clinical stage pharmaceutical company seeking to develop solutions for severe pain relief while reducing the potential for opioid misuse, abuse and overdose. Its platforms include the Trypsin Activated Abuse Protection (TAAP) platform, an abuse-resistant opioid prodrug technology, and the Multi-Pill Abuse Resistant (MPAR) platform, which combines TAAP prodrugs with nafamostat to limit bioavailability after co-ingestion of multiple doses. Its lead product candidate, PF614, is a TAAP oxycodone prodrug under development for acute or chronic pain. A Phase 1 single ascending dose trial in 64 healthy subjects showed that PF614 was well tolerated with no serious adverse events. The PF614-MPAR combination was granted Breakthrough Therapy designation by the FDA in January 2024. A second study, PF614-MPAR-102, designed to evaluate overdose protection across a range of dosages, began enrolling in December 2024. The company describes its primary focus as opioid pain products and opioid use disorder products, and says it has three clinical-stage product candidates.
