TScan Therapeutics, Inc. (Nasdaq: [TCRX](https://aibio.space/companies/tscan-therapeutics/)) announced on November 3, 2025 a workforce reduction of about 30%, or 66 roles, as part of a strategy to concentrate on its blood cancer program. It reported the restructuring in a [Form 8-K](https://www.sec.gov/Archives/edgar/data/1783328/000119312525261612/d873161d8k.htm) under Item 2.05, which covers costs tied to exit or disposal activities.

## What the filing says

The company calls the plan a strategic prioritization. It will concentrate clinical development on its heme program, pause further enrollment in its solid tumor Phase 1 trial, and focus preclinical work on in vivo engineering for solid tumors and on target discovery in autoimmunity. The workforce reduction is part of that plan.

TScan expects to record a one-time charge of up to approximately $2.3 million in the quarter ending December 31, 2025, for severance-related benefits and other costs. It warns that actual results may differ materially. The plan is expected to produce annual cost savings of $45.0 million in 2026 and 2027.

Chief Financial Officer Jason A. Amello was quoted in the company's press release, furnished with the 8-K. "By focusing clinical development on heme and rightsizing the organization accordingly across all functions, we expect to realize annual cost savings of approximately $45.0 million in 2026 and 2027," he said. He added that the company has extended its cash runway into the second half of 2027.

## The TSC-101 trial plan

The 8-K says the company met with the FDA in October 2025 about a pivotal trial design for TSC-101. The product is meant to treat residual disease and prevent relapse in patients with AML or MDS who undergo allogeneic hematopoietic cell transplantation with reduced intensity conditioning. The FDA agreed to a study design that mirrors the current Phase 1 ALLOHA trial.

The updated design uses a biologically assigned internal control arm instead of an external control arm drawn from the CIBMTR registry. The company plans to dose about five more patients at this dose level before starting the pivotal trial, which it now expects to begin in the second quarter of 2026.

The company also disclosed that it recently observed relapse or prolonged incomplete chimerism, measured with a high sensitivity assay, in some TSC-101 patients enrolled in the Phase 1 study in 2025. It says these cases appear to be linked to products that had higher levels of T cell expansion in manufacturing. A new commercial-ready process takes 12 days instead of 17 days and requires less T cell expansion. Data from the ALLOHA trial, including two-year relapse data on the first patients, will be presented at the American Society of Hematology meeting on December 6, 2025.

It plans two more investigational new drug applications in the fourth quarter of 2025 to expand HLA coverage, with Phase 1 development targeted for the second half of 2026 subject to additional funding.

## Solid tumor and autoimmunity programs

In the PLEXI-T solid tumor trial, the first two patients were dosed in October 2025 with multiplex TCR-T therapy candidates. Seven patients have been treated to date with singleplex TCR-T at dose level 3 or higher. The company is pausing further enrollment and says it has recently partnered with a third party that specializes in a lentiviral-based platform for in vivo engineering of T cells. It expects to share initial safety and efficacy data from PLEXI-T in the first quarter of 2026.

In autoimmunity, the company is continuing to discover targets for Crohn's disease in partnership with Amgen. The 8-K moves the solid tumor work to preclinical in vivo engineering.

## About the company

In its annual report on Form 10-K filed March 5, 2025, TScan describes itself as a clinical-stage biotechnology company developing T cell receptor (TCR)-engineered T cell therapies for patients with cancer.

## Losses behind the cut

The 10-K shows a net loss of $127,499 thousand for 2024, against $89,218 thousand for 2023, and cash, cash equivalents and restricted cash of $183,720 thousand at the end of 2024. The 10-K also lists among its risks that raising additional capital may dilute existing stockholders. The company says its cash runway now extends into the second half of 2027. That is the company's own projection, and the 8-K does not give a cash balance at the date of the cut.

Gavin MacBeath signed the 8-K as Chief Executive Officer.
