Sagimet Biosciences Inc. filed an investor presentation with a Form 8-K on October 22, 2026. The company says its $257.6 million in cash, cash equivalents and marketable securities at June 30, 2026, together with proceeds from a $115 million underwritten offering of Series A Common Stock completed in September 2026, is expected to fund current operations through mid-2029. The company says that period includes the readout of the denifanstat Phase 3 acne trial. The presentation does not name the underwriters, the offering price or the number of shares sold.

## Cash and losses

XBRL data from the annual report filed March 11, 2026 show a 2025 net loss of $51.0 million, up from $45.6 million in 2024. Research and development expense was $39.1 million in 2025 and $38.4 million in 2024. Cash used in operations was $45.6 million in 2025, against $42.4 million in 2024. The same data show year-end 2025 cash and cash equivalents of $35.0 million and year-end short-term investments of $78.1 million, as reported in a 10-Q filed August 11, 2026. Cash and cash equivalents were $165.9 million at June 30, 2026, according to that 10-Q. The presentation's $257.6 million figure covers cash, cash equivalents and marketable securities together.

## The acne program

Denifanstat is an oral, once-daily FASN inhibitor. The company is developing it for acne, for MASH, a form of metabolic liver disease, and for select cancers. The presentation says denifanstat met all primary and secondary endpoints in a Phase 3 acne trial, called ASC40-303. That trial was run in China by Ascletis, the company's license partner for Greater China, which announced that a new drug application was accepted for review by China's NMPA in December 2025. The presentation cites Ascletis data on file for 480 patients randomized 1:1 to denifanstat 50mg or placebo for 12 weeks. Treatment success on the investigator global assessment was 33.2 for denifanstat and 14.6 for placebo, a placebo-adjusted 18.6. Dry eye was reported in 10.9% of denifanstat patients and 8.0% of placebo patients, and dry skin in 6.3% and 2.9%. The presentation reports no denifanstat-related grade 3 or 4 adverse events. The data come from the partner, not from Sagimet.

The U.S. trial, called AURORA, dosed its first patient in October 2026, after an FDA "Study May Proceed" letter in July 2026. It is a double-blind, placebo-controlled study of 800 patients, with 533 assigned to denifanstat 50mg and 267 to placebo. The co-primary endpoints at week 12 are the percentage of patients who achieve treatment success, and the absolute change in inflammatory and non-inflammatory lesion counts. The presentation says the study includes 450 patients aged 12 to 17. The company expects topline results in 2H 2027, which falls before the mid-2029 date the company gives for its cash runway.

## The MASH evidence

The MASH data are older and smaller. The Phase 2b FASCINATE-2 trial enrolled 168 patients with stage F2 or F3 fibrosis and ran for 52 weeks. In the F3 modified intention-to-treat population, fibrosis improved by at least one stage with no worsening of MASH in 49% of denifanstat patients and 13% of placebo patients. In the modified intention-to-treat population, progression to cirrhosis was 5% on denifanstat and 11% on placebo, a difference the company describes as statistically significant. Adverse events leading to treatment discontinuation occurred in 19.6% of the denifanstat group and 5.4% of the placebo group.

The annual report said the company planned a Phase 2 proof-of-concept trial of a denifanstat and resmetirom combination in patients with F4 fibrosis, expected to start in the second half of 2026, subject to consultation with regulatory authorities. The presentation says further MASH development will be undertaken only upon securing non-dilutive funding.

## The pipeline and patents

The company's second FASN inhibitor, TVB-3567, received Investigational New Drug clearance in March 2025 and started a first-in-human Phase 1 trial in June 2025. The presentation expects a Phase 2 proof-of-concept trial to begin before the end of 2026. Denifanstat's composition-of-matter patent is listed to expire in 2032, with a potential extension to 2037. TVB-3567's is listed to expire in 2035, with a potential extension to 2038.

The presentation does not break the $115 million out by program.
