On July 30, 2026, PTC Therapeutics, Inc. reported a corporate update and financial results for the second quarter ended June 30, 2026. Total revenue was $361 million, including $239 million of product revenue. Sephience (sepiapterin) contributed $151 million of second quarter revenue, and the company said 1,647 patients were on commercial therapy worldwide as of June 30, 2026. The results were furnished in an [8-K](https://www.sec.gov/Archives/edgar/data/1070081/000107008126000016/tmb-20260728xex99d1.htm).

## Revenue and guidance

Net product revenue across the commercial portfolio was $238.8 million for the second quarter, compared with $118.3 million a year earlier. Sephience net product revenue was $151.3 million, up 21% from the first quarter of 2026. Translarna net product revenue was $42.2 million, compared with $59.5 million a year earlier. Emflaza net product revenue was $24.6 million, compared with $36.4 million, which the company attributed to continued generic erosion. Both Duchenne products fell year on year. Royalty revenue from Roche's sales of Evrysdi (risdiplam) was $71.1 million for the quarter. Net product and royalty revenue totaled $309.9 million, compared with $175.9 million a year earlier.

PTC also recorded a $50.0 million development milestone from Novartis for the first patient dosed in the Phase 3 INVEST-HD study, which was recorded as collaboration revenue. Net income was $83.5 million for the quarter, compared with a net loss of $64.8 million for the second quarter of 2025. The $50.0 million milestone is a one-time payment and helped that result.

For 2026, the company now expects total revenue of $1.18 to $1.28 billion, and total product revenue of $850 to $950 million, up from $750 to $850 million. GAAP research and development and selling, general and administrative expense guidance remains $775 to $815 million. Non-GAAP guidance remains $680 to $720 million, excluding estimated non-cash, stock-based compensation expense of $95 million.

## Pipeline and corporate updates

Chief Executive Officer Matthew B. Klein, M.D., said, "Our teams delivered another strong quarter enabling us to again raise 2026 product revenue guidance." The company also named PTC303 as its next small molecule splicing development candidate, targeting Huntington's disease and myotonic dystrophy, with a plan to enter the clinic in 2027.

For votoplam, PTC reported 24-month PIVOT-HD results showing dose-dependent slowing on the cUHDRS scale in Stage 2 participants, reaching 52% at the 10 mg dose level. The release gives no p-values, patient numbers or control comparison, and the Phase 3 trial that Novartis runs has only begun enrolling. The FDA has not yet weighed in. The company plans to engage with the FDA in the second half of 2026. The Phase 3 INVEST-HD study, which Novartis conducts and funds, is targeting about 770 individuals with early symptomatic disease.

The PROVE-FA study of vatiquinone is expected to start in the third quarter of 2026 to support an NDA resubmission for Friedreich's ataxia, so vatiquinone is not approved for it. It will enroll about 120 individuals aged 7 to 21 and includes an open-label arm with a matched natural history control group. PTC612, an oral NLRP3 inhibitor, entered Phase 1 in the second quarter, and a Phase 2a study of PTC844, a next-generation DHODH inhibitor, is expected to start in the third quarter.

## Balance sheet and financing

GAAP research and development expense was $99.2 million for the quarter, compared with $113.0 million a year earlier. GAAP selling, general and administrative expense was $80.6 million, compared with $85.3 million. In June 2026, PTC issued $550.0 million of senior convertible notes due in 2031 at a 0% coupon, with a conversion price representing a 40% premium over the stock's closing price at the time of the deal. Concurrently, it repurchased the majority of its 1.5% senior convertible notes due in September 2026. The new notes carry no coupon but can convert into shares, which would dilute holders.

Cash, cash equivalents and marketable securities were $2,229.1 million on June 30, 2026, compared with $1,945.4 million on December 31, 2025. The company had 83,327,286 shares issued and outstanding on June 30, 2026.

## Background

In its annual report, PTC describes itself as a global biopharmaceutical company focused on rare disorders in neurology and metabolism. The 8-K's guidance and pipeline updates build on products already on the market, including Sephience, Translarna, Emflaza, Kebilidi and Evrysdi.
