On May 27, 2026, MetaVia Inc. (Nasdaq: [MTVA](https://aibio.space/companies/metavia/)) presented new Phase 1 data on DA-1726 in a late-breaking poster at the European Association for the Study of the Liver Congress 2026 in Barcelona, Spain. The congress runs May 27 to 30, 2026. The company's release reports results from the 48 mg cohort of a randomized, double-blind, placebo-controlled Phase 1 multiple ascending dose study in obese but otherwise healthy adults.

## Study design

Subjects received once-weekly subcutaneous DA-1726 or placebo for four weeks, without dose titration, in a 2:1 randomization ratio. Of the nine subjects enrolled in the 48 mg cohort, six entered an optional four-week extension phase at the same dose. Chris Fang, Chief Medical Officer, is the presenting author.

## Weight and waist circumference

The mean body-weight reduction was 6.1% at Day 26 and 9.1% at Day 54, with no evidence of a plateau through Week 8. Waist circumference was reduced by 5.8 cm at Day 26 and by 9.8 cm at Day 54. The release describes these as clinically meaningful reductions in body weight and waist circumference, but the version we have does not show the statistics behind them. Chief Executive Officer Hyung Heon Kim said the company saw "robust and progressive weight loss" at the 48 mg dose level.

## Safety and tolerability

DA-1726 was generally well tolerated up to the 48 mg dose level. The release reports no serious adverse events and no treatment-related discontinuations. Gastrointestinal adverse events were mostly mild to moderate and transient, even without titration. Heart rate and QTcF showed no clinically meaningful changes, despite activation of the glucagon receptor. The annual report, however, says the Part 3 titration studies aim to remove moderate adverse effects seen at the 48 mg dose level.

## Exploratory liver measures

The company described its noninvasive liver assessments using FibroScan as exploratory. At Day 54, controlled attenuation parameter (CAP) fell by 20.0 dB/m in the 48 mg group, compared with a rise of 24.0 dB/m in the placebo group. Liver stiffness changed by minus 10.3% from baseline by vibration-controlled transient elastography, compared with plus 13.8% with placebo. The release also reports directional improvements from baseline in the FibroScan-aspartate aminotransferase (FAST) score, which the company presented alongside the stiffness and CAP results. The group is small, nine subjects in the cohort, and these are obese but otherwise healthy adults rather than patients with liver disease, so the liver readings are a first signal at best. The company says these findings support further long-term evaluation of DA-1726 in obesity-associated liver disease and MASH. The company says the findings further illustrate the potential of DA-1726 in obesity and MASH, where metabolic and hepatic dysfunction are closely linked.

## The program and the company

DA-1726 is a novel oxyntomodulin analogue that works as a GLP1R and GCGR dual agonist. The company is developing it for obesity and MASH, with once-weekly subcutaneous dosing. In its annual report, MetaVia said the Phase 1 trial was its first-in-human study for DA-1726. Ongoing Phase 1 Part 3a and Part 3b titration studies continue to evaluate extended treatment at higher dose levels. The annual report planned the data readout for both parts in the fourth quarter of 2026.

MetaVia's second asset, vanoglipel (DA-1241), is a GPR119 agonist in development for MASH and type 2 diabetes. The annual report describes four legacy programs that the company is not planning to advance.

## Financial position

The annual report, filed March 26, 2026, shows research and development expense of $6.8 million for 2025, compared with $21.6 million for 2024. Net loss was $13.0 million for 2025 and $27.6 million for 2024. Cash from operations was negative $15.7 million for 2025 and negative $24.7 million for 2024. The release's own risk list names the company's history of net losses and the sufficiency of its cash on hand to fund operations.
