## The restructuring

Korro Bio, Inc. (Nasdaq: [KRRO](https://aibio.space/companies/korro-bio/)) said on November 12, 2025 that it is carrying out a strategic restructuring that cuts its workforce by about 34%, across all levels of the organization. The company says the cuts extend its cash runway into the second half of 2027. It estimates one-time restructuring charges of about $2.4 million for severance, benefits and related termination costs, most of which it expects to record in the fourth quarter of 2025.

The chief executive, Ram Aiyar, said the cuts are meant to focus resources on generating clinical data and advancing GalNAc-conjugated programs that target the liver. The release also says Chief Medical Officer Dr. Kemi Olugemo resigned effective November 12, 2025.

## What happened to KRRO-110

The restructuring follows a disappointing update on KRRO-110, the company's most advanced program in alpha-1 antitrypsin deficiency, or AATD. Korro said the drug produced functional protein in AATD patients after a single administration, but did not reach the protein levels it projected. The release gives the greatest peak total AAT protein as about 10 µM, against a protective threshold of 11 µM. The greatest increase in functional M-AAT protein from baseline was about 2 µM, and it lasted up to four weeks in the first patient evaluable by LC/MS.

The trial is the Phase 1/2a REWRITE study. Korro has completed all six planned single-ascending-dose cohorts in healthy volunteers, with 24 participants, and seven AATD patients have been dosed in two open-label cohorts at 0.6 mg/kg and 0.8 mg/kg. The release says there are no plans for more single-dose AATD cohorts, and that the company is still weighing next steps for the multiple-dose portion. Mild-to-moderate infusion-related reactions were seen in two healthy volunteers at 1.2 mg/kg and in four AATD patients, and all resolved within 24 hours. The release reports no dose-limiting toxicities or treatment-emergent serious adverse events as of a November 6, 2025 data cutoff. Korro says the safety profile is consistent with lipid nanoparticle infusion-related class effects.

The company also says initial analysis shows pharmacokinetic differences between healthy volunteers and AATD patients, with apparently faster dissociation of the drug in patients. It cites those findings among the insights behind its pivot to a GalNAc-conjugated construct, with a development candidate nomination expected in the first half of 2026. The release does not say whether the new construct will reach the protein levels the original design missed.

## Cash, losses and the Novo Nordisk pause

Korro had $102.5 million in cash, cash equivalents and marketable securities at September 30, 2025, down from $163.1 million at December 31, 2024. It says that balance will fund operating expenses and capital spending into the second half of 2027.

In the third quarter, Korro recorded collaboration revenue of $1.1 million, from its Novo Nordisk collaboration, against none a year earlier. Research and development expense was $13.8 million, down from $16.0 million, and general and administrative expense was $6.5 million, down from $7.3 million. The net loss was $18.1 million for the quarter, compared with $21.0 million a year earlier.

The Novo Nordisk amendment sets a 12-month pause to reassess the rationale for the current target under the first research program. The release does not say whether the pause changes any payments under the collaboration. Korro has also nominated a second development candidate, KRRO-121, for hyperammonemia, including urea cycle disorders and hepatic encephalopathy. It expects to file to start a first-in-human trial in the second half of 2026.

## The earlier claims

The annual report filed March 18, 2025, before the update, described KRRO-110's "best-in-class potential" for AATD. The November release does not explain the gap between that description and the protein levels it reports. The release's forward-looking statements warn that the timing and results of trials could differ from expectations, and they list the cash runway among the items at risk. It does not say what the restructuring will cost beyond the $2.4 million charge.
