Kiniksa Pharmaceuticals International, plc (Nasdaq: [KNSA](https://aibio.space/companies/kiniksa-pharmaceuticals-international/)) said on February 25, 2025 that it is discontinuing its Phase 2b clinical trial of abiprubart in Sjögren's Disease and ending its license for mavrilimumab with MedImmune, Limited. The company disclosed both steps in a [Form 8-K](https://www.sec.gov/Archives/edgar/data/1730430/000110465925016725/tm257489d1_8k.htm). The 8-K does not report any efficacy results from the trial.

## The abiprubart trial wind-down

On February 24, 2025, citing its strategic reprioritization and capital allocation, Kiniksa committed to a plan to discontinue the Phase 2b trial. It expects to end enrollment immediately, with winddown activities completed by the end of 2025. The company has incurred approximately $19 million in expenses so far and expects to record approximately $14 million to $17 million more.

Almost all of the additional cost is contract termination fees under the clinical supply agreements for abiprubart, plus small trial closeout costs. The company plans to book the charges as research and development expenses, mostly in the fourth quarter of 2024 and the first half of 2025. The 8-K says the estimates rest on assumptions and that actual results may differ materially. The company says it will explore strategic alternatives for the asset. The filing does not name a buyer or partner for it.

In the press release, Chairman and Chief Executive Officer Sanj K. Patel said: "In line with our prioritization of cardiovascular indications, we plan to discontinue the development of abiprubart in Sjögren's Disease."

## The MedImmune license

On February 21, 2025, a wholly owned subsidiary gave MedImmune, Limited written notice that it is terminating the license agreement dated December 21, 2017, as amended on July 9, 2020. The company exercised its right to terminate for convenience, and the termination takes effect on May 22, 2025. The license gave Kiniksa an exclusive, sublicensable, worldwide right to develop and commercialize mavrilimumab, an antibody to the GM-CSF receptor alpha, for all indications. That license ends on the termination date.

The filing is therefore a retreat on two fronts: the company is dropping a clinical program and giving up the rights to a second asset it had licensed.

## The money

Kiniksa reported revenue of $423.2 million for 2024, up from $270.3 million for 2023. ARCALYST net product revenue was $417.0 million for the year. Total operating expenses were $468.9 million, and the company reported a net loss of $43.2 million, compared with net income of $14.1 million in 2023. The company had $243.6 million of cash, cash equivalents and short-term investments and no debt at December 31, 2024, up from $206.4 million a year earlier.

The net loss came in 2024 despite the revenue growth, and the release says tax benefits affected the quarterly and annual results. The company expects 2025 ARCALYST net product revenue of between $560 million and $580 million, and says its current operating plan will remain cash flow positive on an annual basis. Those figures are company guidance, not results.

## The pipeline that remains

Kiniksa says it is focusing on diseases with unmet need and prioritizing cardiovascular indications. Its new program, KPL-387, is an investigational monthly antibody for recurrent pericarditis. The press release says Kiniksa is running a Phase 1 single and multiple ascending dose study of KPL-387, and plans a Phase 2/3 trial in mid-2025, with Phase 2 data expected in the second half of 2026.

KPL-387 is in Phase 1 today. The Sjögren's program the company is ending had reached Phase 2b. The 8-K was signed by Madelyn Zeylikman, Senior Vice President, General Counsel and Secretary.
