Alector, Inc. (Nasdaq: [ALEC](https://aibio.space/companies/alector/)) said on October 21, 2025 that it will cut about 49% of its workforce after its Phase 3 INFRONT-3 trial of latozinemab in frontotemporal dementia missed its clinical co-primary endpoint. The company reported the restructuring in a [Form 8-K filed October 21, 2025](https://www.sec.gov/Archives/edgar/data/1653087/000119312525245102/alec-20251020.htm) under Items 2.02, 2.05, 5.02 and 7.01. Arnon Rosenthal, Ph.D., Alector's chief executive officer, signed the filing.

## The trial result

INFRONT-3 was a pivotal, randomized, double-blind, placebo-controlled trial of latozinemab, developed with GSK, in people with frontotemporal dementia caused by a progranulin gene mutation. Patients received 60 mg/kg intravenously every four weeks for 96 weeks. The primary analysis covered symptomatic participants only.

The trial did not meet the clinical co-primary endpoint of slowing disease progression, measured by the Clinical Dementia Rating plus the NACC FTLD-SB scale. The company says treatment did produce a statistically significant effect on plasma progranulin concentrations, the biomarker co-primary endpoint. That biomarker was added at the U.S. Food and Drug Administration's recommendation, through an amended statistical analysis plan. The release says the designation of plasma progranulin as a co-primary endpoint does not apply outside the United States.

The secondary and exploratory endpoints, including fluid biomarkers and volumetric MRI, showed no treatment-related effects. Alector says preliminary safety data have not highlighted any major safety concerns, but that a more detailed analysis is ongoing. Chief Medical Officer Giacomo Salvadore, M.D. said in the release that latozinemab "did not demonstrate a clinical benefit in INFRONT-3," and that the insights gained are invaluable. The results will be presented at an upcoming medical congress.

Because of these results, the open-label extension and the continuation study for latozinemab will be discontinued.

## The restructuring

The company committed to the plan on October 21, 2025 to align resources with its strategic priorities. The reduction in force affects about 75 employees across the organization. Alector expects total incremental restructuring charges of about $7.7 million, covering severance and related termination benefits. The reduction is expected to be completed during the first half of 2026.

## The cash figure

Alector estimates it had approximately $291.1 million in cash, cash equivalents and short-term investments as of September 30, 2025. The company says that amount should provide runway through 2027. The 8-K calls the figure a preliminary estimate. The company plans to issue 2026 guidance when it reports fourth-quarter and full-year 2025 results. The filing does not give a net loss for the period, so the size of the cash burn behind the runway claim cannot be measured from it.

## The departure

Sara Kenkare-Mitra, Ph.D., President and Head of Research and Development, informed the company on October 20, 2025 that she will resign effective December 22, 2025. The 8-K says the resignation is not the result of any disagreement with the company over its operations, policies or practices. She will stay until then to support a transition across the R&D organization.

The company has agreed to a separation agreement with her. It provides a lump-sum payment equal to nine months of her annual base salary, a payment equal to 50% of her annual bonus target, and nine months of company-paid COBRA premiums, in exchange for a customary general release of claims. Alector says it intends to file the agreement as an exhibit to a later periodic report.

## The pipeline

The company says it remains focused on neurodegenerative diseases. Nivisnebart, also called AL101 or GSK4527226, is developed in a collaboration with GSK. It is in PROGRESS-AD, a 76-week Phase 2 trial in early Alzheimer's disease. Enrollment finished in April 2025, trial completion is expected in 2026, and an independent interim analysis is planned for the first half of 2026.

Alector's other pipeline work relies on its Alector Brain Carrier platform, which it describes as a blood-brain barrier technology. The company is targeting investigational new drug applications for AL137, an anti-amyloid beta antibody for Alzheimer's, in 2026, and for AL050, a glucocerebrosidase enzyme replacement for Parkinson's disease, in 2027.

The 8-K ties the cuts to the trial result and to the company's strategic priorities. The release gives no date for another pipeline readout beyond the planned PROGRESS-AD interim analysis.
