Alaunos Therapeutics, Inc. (Nasdaq: [TCRT](https://aibio.space/companies/alaunos-therapeutics/)) said on June 29, 2026 that a new preclinical statistical analysis of liver weight, adjusted for body fat percentage, supports a lower liver weight signal for ALN1003, its investigational oral small-molecule candidate for obesity and metabolic disorders. The analysis draws on a 48-day diet-induced obesity (DIO) mouse study that the company had reported earlier on a non-Good Laboratory Practice (non-GLP) basis. Alaunos described the result as supportive evidence for further controlled preclinical work, not as proof of a mechanism.

## The new analysis

In that study, mice treated with ALN1003 had lower liver weight than controls after adjustment for body fat percentage, using a standard ANCOVA analysis (p=0.000005). The same conclusion held when the company used heteroscedasticity-robust HC3 standard errors as a sensitivity analysis (p=0.000015). The company says the finding indicates that the lower liver weight was not fully explained by measured body fat in this model. The release also says the analysis is post hoc and that its p-values are nominal, so the very small p-values should not be read as confirmed results.

The release places the result beside earlier findings from the same study. A previously reported blinded pilot pathology review of selected liver samples showed qualitative findings consistent with lower hepatic steatosis, with a mean NAS score of 2.7 for treated animals compared with 5.0 for selected controls, so the samples were not a full set. That earlier work also showed a 43% reduction in absolute liver weight and significantly lower liver injury markers (ALT, AST and ALP). Alaunos stressed that these limited pilot findings do not establish MASLD resolution, fibrosis reversal, inflammation improvement or clinical efficacy.

Alaunos has also reported lower fasting insulin and lower HOMA-IR, a calculated fasting glucose and insulin index. The HOMA-IR finding stayed statistically significant after adjustment for percentage body fat (ANCOVA p=0.0006; HC3 robust sensitivity p=0.0014). Holger Weis, the company's chief executive, said: "This analysis adds an important supportive piece to the ALN1003 preclinical dataset."

## The program and the financial record

The 10-K for 2025 describes Alaunos as a preclinical-stage biopharmaceutical company focused on orally administered small-molecule therapeutics for obesity and related metabolic disorders, including MASLD. The filing says the company was previously a clinical-stage, oncology-focused cell therapy company developing adoptive TCR-T cell therapies. ALN1003 is being evaluated for potential relevance to multiple components of metabolic dysfunction. The release lists limited sample sizes and single-timepoint biomarker assessments among its limitations. ALN1003 has not been evaluated in human clinical trials.

Alaunos reported a net loss of $4.2 million for 2025, compared with $4.7 million for 2024. Research and development expense rose to $1.4 million in 2025 from $362,000 in 2024. Cash used in operations was $2.9 million in 2025 and $5.0 million in 2024, and year-end cash and cash equivalents for 2024 were $1.1 million. The accumulated deficit was $924.6 million as of December 31, 2025.

The release says the company had cash and cash equivalents of approximately $0.354 million as of March 31, 2026, and that it intends to pursue additional financing to support continued operations and the preclinical program. It does not state a cash runway.

## Limits and what is still needed

The release gives no human data, and the 10-K says the first study used 12 mice per group. Alaunos said the body-fat-adjusted result supports further controlled preclinical evaluation of ALN1003's liver-related effects. Its chief executive named formulation, exposure-response, tolerability and MASH-relevant liver biology as areas for further controlled studies, and said the findings support continued development of the compound.

The company points to an investor presentation from May 2026 that holds the data summaries and liver histology images. Mouse results of this kind often do not carry over to people. With $0.354 million of cash at March 31, 2026 and no stated runway, the company needs new financing before it can pay for the controlled studies it describes.
