# Takeda bets its rebound on three launches as profit dips

Orzeyful and Mimrylo are approved and zasocitinib is under priority review. Core operating profit is guided to fall 5% to 8% this year.

Takeda is in the middle of a planned dip. Revenue in the year to March 2026 [fell 1.7% at actual rates](https://www.takeda.com/newsroom/newsreleases/2026/takeda-reports-fourth-quarter-fy2025-results/) after the ADHD drug Vyvanse lost exclusivity, and guidance for the current year calls for core operating profit to fall 5% to 8% at constant rates. The company's answer is three new medicines, all approved or under FDA review in 2026, and a new CEO, Julie Kim, who took over in June.

## The technology

Takeda is a diversified drugmaker with businesses in gastrointestinal and inflammation, rare diseases, plasma-derived therapies, oncology, neuroscience and vaccines. Entyvio is its largest product, with [first-quarter sales of JPY 268.3 billion](https://www.sec.gov/Archives/edgar/data/1395064/000139506426000327/form6k_073026.htm). Plasma-derived immunoglobulins added JPY 213.9 billion.

The new wave is small-molecule and peptide drugs aimed at specific biology. Orzeyful is the first orexin receptor 2 agonist approved for narcolepsy type 1, [cleared by the FDA on August 5](https://www.takeda.com/newsroom/newsreleases/2026/orzeyful-approved-narcolepsy/). Mimrylo is a hepcidin mimetic [approved on August 28](https://www.takeda.com/newsroom/newsreleases/2026/fda-approval-mimrylo/) for polycythemia vera. Zasocitinib is a TYK2 inhibitor that [beat deucravacitinib head to head](https://www.takeda.com/newsroom/newsreleases/2026/zasocitinib-outperforms-deucravacitinib-study/) in psoriasis. Behind them sit two oncology assets licensed from Innovent, a [PD-1/IL-2 bispecific called TAK-928](https://www.takeda.com/newsroom/newsreleases/2026/esmo-pipeline-progress/) in a global lung cancer phase 3, and the Claudin 18.2 antibody-drug conjugate arcotatug tavatecan, which has [late-breaking phase 3 data at ESMO](https://www.takeda.com/newsroom/newsreleases/2026/esmo-pipeline-progress/).

## The odds

Two of the three launches are already approved. Zasocitinib is under [FDA priority review](https://www.takeda.com/newsroom/newsreleases/2026/fda-priority-review-zasocitinib-psoriasis/) with a decision due in the first quarter of 2027. In the [BIO/Informa/QLS 2011-2020 analysis](https://go.bio.org/rs/490-EHZ-999/images/ClinicalDevelopmentSuccessRates2011_2020.pdf), 90.6% of filed applications were approved. With two positive registrational trials and a head-to-head win, zasocitinib looks closer to that base rate than most filings.

The harder odds are commercial. Takeda's own guidance shows the new products made up only [4% of core revenue](https://www.sec.gov/Archives/edgar/data/1395064/000139506426000327/form6k_073026.htm) in the first quarter. Vyvanse fell 17.0% and Takhzyro 2.2% at constant rates, the latter on US competition.

## The money

Takeda is profitable but carries substantial debt. Net debt was [JPY 4,479.0 billion at June 30](https://www.sec.gov/Archives/edgar/data/1395064/000139506426000327/exhibit991_073026.htm), with bonds and loans of JPY 4,940.0 billion and cash of JPY 461.0 billion. Adjusted net debt was 2.7 times adjusted EBITDA. Adjusted free cash flow was JPY 684.5 billion in fiscal 2025 and JPY 68.6 billion in the first quarter. The fiscal 2026 dividend forecast is [JPY 204 per share](https://www.takeda.com/newsroom/newsreleases/2026/financial-results-update-amitiza-antitrust-litigation/).

## The risks

The Amitiza antitrust verdict is the largest known liability. A US jury awarded about [USD 885 million in single damages](https://www.takeda.com/newsroom/newsreleases/2026/takeda-provides-update-regarding-jury-verdict-in-amitiza-antitrust-litigation-in-the-us-and-related-revision-to-fy2025-financial-results/), most of it subject to trebling, and Takeda booked a JPY 402.5 billion provision while it appeals.

Launch execution is the second risk. Orzeyful launched in Japan and China on October 15 and is [set to launch in the US in November](https://www.takeda.com/newsroom/newsreleases/2026/oveporexton-launches-simultaneously-in-japan-and-china/), following DEA scheduling. Its US label lists insomnia in more than half of patients. Leadership is also changing: R&D president Andy Plump will [retire in June 2027](https://www.takeda.com/newsroom/newsreleases/2026/planned-retirement-of-andy-plump/).

## Our read

Takeda has done the hard part of the pipeline work. Three drugs with first-in-class or best-in-class claims reached approval or filing within a few months. The question now is commercial. They need to scale fast enough to offset Vyvanse and Entyvio's slowing growth, while debt and a large legal provision limit room for deals.

Kim's [capital markets day on December 11](https://www.takeda.com/newsroom/newsreleases/2026/takeda-reports-first-quarter-fy2026-results/) is the moment to judge the plan in detail, including how Takeda splits cash between launches, debt reduction and the Amitiza appeal, and who replaces Plump as head of R&D. Before that, second-quarter results on [October 29](https://www.takeda.com/newsroom/newsreleases/2026/earnings-call-q2-fy2026/) cover the period to September 30, before Orzeyful's October 15 launch in Japan and China, and the zasocitinib decision follows in early 2027.
