# Revolution Medicines turns a RAS target into an approved drug

Rasonque won FDA approval 35 days after the filing was accepted. The lung programs are next, and the company holds about $3.9 billion to fund them.

## The technology

Revolution Medicines makes small molecules that block RAS, a family of proteins that drive a large share of pancreatic, lung and colorectal cancers. Drug makers spent decades unable to hit RAS. Revolution's drugs are built to hit the active form of RAS, which the company calls RAS(ON). The company calls its lead drug, daraxonrasib, a [RAS(ON) multi-selective inhibitor](https://ir.revmed.com/news-releases/news-release-details/revolution-medicines-new-drug-application-daraxonrasib-accepted), which means it is built to work across several RAS variants, not one mutation.

That breadth is the commercial point. In RASolute 302, patients with a wide range of RAS variants and wild-type tumors were enrolled, and the [FDA label requires no companion diagnostic](https://www.pharmaceutical-tech.com/news/fda-approves-revolution-medicines-rasonque). The pipeline extends the idea: zoldonrasib and elironrasib for lung cancer, RMC-5127 aimed at G12V, and RM-055, a preclinical molecule meant to address resistance, per the [Fierce Biotech report](https://www.fiercebiotech.com/biotech/aacr-revolution-rolls-pancreatic-cancer-ras-wave-rises) from AACR.

## The odds

Daraxonrasib is already approved, so the question for the lead drug is commercial, not regulatory. For the rest of the pipeline, base rates apply. The BIO, Informa and QLS analysis of 2011 to 2020 found that [28.9% of programs move from phase 2 to phase 3, and 57.8% of phase 3 programs reach an NDA or BLA](https://go.bio.org/rs/490-EHZ-999/images/ClinicalDevelopmentSuccessRates2011_2020.pdf). The likelihood of approval from phase 3 across all indications was 52.4%.

Revolution has better-than-average evidence for its lead drug. [RASolute 302 randomized 500 patients](https://www.biospace.com/press-releases/revolution-medicines-announces-asco-plenary-presentation-highlighting-unprecedented-results-from-pivotal-phase-3-rasolute-302-clinical-trial-of-daraxonrasib-in-previously-treated-metastatic-pancreatic-cancer) and found a hazard ratio of 0.40 for overall survival. The lung studies have less to go on. The zoldonrasib and elironrasib first-line data come from [28 and 39 patients](https://www.sec.gov/Archives/edgar/data/0001628171/000119312526335039/rvmd-ex99_1.htm) in single-arm studies with a May 11 cutoff. These need randomized trials. Daraxonrasib's lung trial, RASolve 301, expects its first readout in 2027.

## The money

Revolution held about [$3.94 billion in cash and marketable securities](https://www.sec.gov/Archives/edgar/data/0001628171/000119312526335039/rvmd-ex99_1.htm) on June 30, up from $2.03 billion at the end of 2025. The increase came from $2.225 billion of gross proceeds in April (stock plus 0.50% convertible notes due 2033) and a $250.0 million Royalty Pharma payment in May. Royalty Pharma committed up to $1.5 billion more, subject to milestones.

Spending is climbing. The second-quarter net loss was $644.4 million, including a $151.0 million non-cash warrant charge, and 2026 GAAP operating expenses are guided to $2.1 billion to $2.2 billion. The release gave no runway figure. At those spending levels the cash covers well over a year, which is our arithmetic and not company guidance, and further phase 3 lung trials plus a commercial launch will push costs up.

The product side is new. Rasonque launched with a [list price of $39,800 for 30 days](https://biopharmadive.com/news/revolution-rasonque-price-pancreatic-cancer-drug-launch-sales/828918/). Evercore ISI models $2.4 billion in pancreatic cancer sales next year, though that is an analyst forecast and not company guidance.

## The risks

Four risks stand out from the filings and press coverage.

The first is execution on the launch. Pancreatic cancer patients are often sick and treated fast, and the payers' response to a price of more than $477,000 a year is untested. Biopharma Dive reported that an RBC analyst expects limited friction but that some insurers [may restrict first-line use because of cost](https://biopharmadive.com/news/revolution-rasonque-price-pancreatic-cancer-drug-launch-sales/828918/).

The second is safety in wider use. Grade 3 or higher rash and stomatitis hit 14% and 12% of patients in the trial, and one patient died of treatment-related pneumonitis, according to the [ASCO release](https://www.biospace.com/press-releases/revolution-medicines-announces-asco-plenary-presentation-highlighting-unprecedented-results-from-pivotal-phase-3-rasolute-302-clinical-trial-of-daraxonrasib-in-previously-treated-metastatic-pancreatic-cancer). The trial had median follow-up of 8.5 months.

The third is trial design. RASolute 302 was open-label and compared against chemotherapy. Overall survival is hard to bias, but progression and response readings are more exposed.

The fourth is competition and dependence. Revolution's value rests heavily on one drug family. [Merck's takeover talks ended](https://www.thepharmaletter.com/pharma-news/merck--co-walks-away-from-revolution-talks-as-price-gap-bites) on price, and a buyer is not assured to return.

## Our read

Revolution has done what most oncology developers do not: it took a hard target to an approval, in a deadly cancer, on a randomized survival win. The phase 3 numbers, a median of 13.2 months against 6.7 months, are large for this disease, and the FDA moved fast under its priority voucher program.

What the market will weigh from here is a different set of problems. The company must show that a high-priced pill sells well, that real-world safety holds, and that the same logic works in lung cancer, where the data are early. The balance sheet gives it room to try. The next dated events are fourth-quarter 2026 items in the [second-quarter release](https://ir.revmed.com/news-releases/news-release-details/revolution-medicines-reports-second-quarter-2026-financial), and the lung readout in 2027.
