# Protagonist has two approvals and a royalty model. The next drug is its own

Icotyde and Mimrylo are now Takeda's and J&J's to sell. Protagonist collects milestones and royalties, buys back stock and bets on an oral IL-17 pill.

## The technology

Protagonist designs peptides, short protein chains, that act like antibodies or hormones but can sometimes be taken by mouth. Two have reached the market. Icotyde (icotrokinra), an oral IL-23 receptor blocker co-discovered with Johnson & Johnson, was [approved in March 2026](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm) for plaque psoriasis in adults and children 12 and older. Mimrylo (rusfertide), a weekly injection that mimics the iron hormone hepcidin, was [approved on August 28](https://www.newswire.com/news/protagonist-therapeutics-announces-u-s-fda-approval-of-hepcidin-mimetic-peptide) for erythrocytosis in polycythemia vera (PV).

The company's own pipeline repeats the formula. PN-881 is an oral IL-17 antagonist that reached [trough levels above its IC90 targets](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm) in a healthy volunteer phase 1 study. PN-477 is a triple GLP-1, GIP and glucagon agonist for obesity, now in phase 1 as an injection. PN-8047 is an oral small-molecule hepcidin mimetic heading into phase 1 in the first quarter of 2027.

## The odds

The two approvals remove most clinical risk from Protagonist's partnered assets. Mimrylo showed a [76.9% response rate versus 32.9%](https://www.newswire.com/news/protagonist-therapeutics-announces-u-s-fda-approval-of-hepcidin-mimetic-peptide) on placebo in phase 3. Icotyde is in phase 3 in psoriatic arthritis, ulcerative colitis and Crohn's disease, run by J&J.

The wholly owned assets are early. Across all diseases, the BIO/Informa/QLS data show phase 1 programs reach phase 2 [52.0% of the time](https://go.bio.org/rs/490-EHZ-999/images/ClinicalDevelopmentSuccessRates2011_2020.pdf) and phase 2 programs reach phase 3 28.9% of the time. PN-881 enters a phase 2b in early 2027 with exposure data only, so efficacy in patients is still unknown.

## The money

Cash was [$849.5 million at June 30](https://www.newswire.com/news/protagonist-therapeutics-announces-300-million-share-repurchase-program), and Takeda owes $275 million more in the fourth quarter. Protagonist posted [net income of $162.8 million](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm) in the second quarter, mostly from the first $200 million Takeda opt-out payment.

Ongoing income comes from royalties. Protagonist gets [6% to 10% on Icotyde](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091123/ptgx-20260630x10q.htm), about 7.25% on average at $4 billion in sales, and 14% to 29% on Mimrylo, about 21% at $1.5 billion. It is also eligible for up to $580 million in J&J milestones and up to [$875 million more](https://www.newswire.com/news/protagonist-therapeutics-announces-u-s-fda-approval-of-hepcidin-mimetic-peptide) from Takeda. In September it authorized a [$300 million buyback](https://www.newswire.com/news/protagonist-therapeutics-announces-300-million-share-repurchase-program).

The company says it does [not anticipate a need for additional funding](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091123/ptgx-20260630x10q.htm) in the near term, though it may raise money later to expand the pipeline or in-license products. Net income in the first half of 2026 was [$166.6 million](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm). Without new partner milestones, reported profits will shrink once the one-time Takeda payments are recognized, and royalties will have to carry the income statement.

## The risks

Protagonist sells nothing itself. Its 10-Q says royalty income depends on [the efforts of our collaboration partner](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091123/ptgx-20260630x10q.htm) and may lag prescriptions early in a launch. Mimrylo carries a list price of about [$218,000 a year](https://pharmaphorum.com/news/fda-clears-takeda-protagonists-first-class-pv-drug), according to Reuters via pharmaphorum, which will need payer coverage.

Spending is rising. The company expects R&D to [increase significantly in the second half of 2026](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm). Obesity is crowded, and PN-477 is still in phase 1. The buyback also competes with pipeline needs for the same cash.

## Our read

Two of Protagonist's molecules became approved drugs within six months of each other. The business is now a royalty holder with a strong balance sheet, which lowers the risk of dilution and funds the next round of science. The open question is whether the platform can deliver a third drug that Protagonist keeps for itself. PN-881's phase 2b, starting in [early 2027](https://www.sec.gov/Archives/edgar/data/1377121/000110465926091099/tm2622272d1_ex99-1.htm), is the first real test. Until then, investors will watch Takeda's and J&J's launch numbers, because those royalties pay for everything else.
