# BioNTech has the cash for oncology, but the proof is still ahead

Pumitamig is in seven late-stage trials, while mRNA programs stumbled in August and October. Revenue guidance was cut and a new CEO is coming.

BioNTech made its name with a COVID-19 vaccine. It is now a cancer company with a falling vaccine business and a large cash balance. The question for investors is whether its oncology bets, led by one bispecific antibody, can turn that cash into approved products before the money is spent.

## The technology

The pipeline has three kinds of drug. The first is next-generation immunomodulators. Pumitamig, developed with Bristol Myers Squibb, binds PD-L1 and VEGF-A, and is in [seven late-stage trials](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000054/a991bntx_q22026pressreleas.htm). Gotistobart targets CTLA-4 in the tumor and is in a phase 3 trial in squamous lung cancer. The second is ADCs: trastuzumab pamirtecan against HER2 and elfetabart drozuntecan against B7-H3, both with Duality Biologics. The third is mRNA cancer immunotherapy, including BNT113 and individualized autogene cevumeran.

BioNTech's stated strategy is to use pumitamig as a backbone and add other agents. Its [ESMO preview](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000069/a991261019_bntxxesmoxcrxpr.htm) lists more than ten such combination trials, and the Gilead deal for a pumitamig and Trodelvy [trial in breast cancer](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000072/a991261022_bntxrosettabrea.htm) extends that idea to a partner's drug.

## The odds

Evidence for pumitamig is still phase 2. In the [ROSETTA Lung-02 interim data](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000039/a99120260530_bntxascoxpumi.htm) from 40 evaluable patients, the confirmed response rate with chemotherapy was 57.1% in non-squamous and 68.4% in squamous lung cancer. Those are response rates, not survival, and the comparison with pembrolizumab plus chemotherapy comes from the phase 3 part now recruiting.

The published base rates suggest caution. In the [BIO, Informa and QLS study of 2011 to 2020](https://go.bio.org/rs/490-EHZ-999/images/ClinicalDevelopmentSuccessRates2011_2020.pdf), 28.9% of programs moved from phase II to phase III, 57.8% from phase III to filing, and 7.9% of programs starting phase I reached approval. BioNTech has had misses of its own in recent weeks: [BNT113 missed its first primary endpoint](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000067/a991261013_bntxbnt113xprim.htm) and the [colorectal autogene cevumeran trial was stopped](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000061/a991260828_bntxxcrcxenxfin.htm). Gotistobart's survival result came from [87 patients](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000063/a991260911_bntxwclcxgotist.htm) in an early first stage.

## The money

BioNTech has €16.6 billion in cash and securities, which is large for a company with first-half revenue of €223.7 million. The [Q2 report](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000054/a991bntx_q22026pressreleas.htm) shows an IFRS net loss of €1.35 billion in the first half. Guidance for 2026 is revenue of €1.6 billion to €1.9 billion, adjusted R&D of €2.0 billion to €2.3 billion and adjusted SG&A of €700 million to €800 million. By simple arithmetic, spending guidance exceeds revenue guidance by roughly €0.8 billion to €1.5 billion, before cost of sales and other items. Third-quarter revenue should include €613 million from Bristol Myers.

The company is also buying back up to $1.0 billion of its ADSs through May 2027, and had spent $151.6 million in the second quarter.

## The risks

Revenue depends on COVID vaccines, where demand is softer than the company expected. Management is changing: [Guido Oelkers replaces Ugur Sahin](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000051/a991biontechannouncesappoi.htm) as CEO by February 1, 2027 at the latest, and chief medical officer Özlem Türeci is leaving to lead a new company. The pipeline leans on one asset, pumitamig, and on one partner, Bristol Myers. And the mRNA franchise, a founding technology for BioNTech, has just had two setbacks.

## Our read

BioNTech is well funded, and its research depth is not in doubt. The weak points are execution and concentration. A phase 3 win for pumitamig in lung cancer would change the company's profile. A loss would leave a large cash balance and a shrinking vaccine business. The next dated checks are the ESMO data and [third-quarter results on November 3](https://www.sec.gov/Archives/edgar/data/1776985/000177698526000054/a991bntx_q22026pressreleas.htm), and later the interim analysis of the gotistobart phase 3 and the primary analysis of the HER2 ADC breast cancer trial in the fourth quarter.
