# Beam's base editing works in the clinic; selling it is the next test

Risto-cel heads toward a BLA and BEAM-302 is in a registrational cohort. The funding plan leans on milestone-linked debt.

## The technology

Beam Therapeutics uses base editing, a form of gene editing designed to correct single-letter changes in DNA. It has two kinds of programs. Risto-cel edits a patient's own blood stem cells outside the body to raise fetal hemoglobin in sickle cell disease. BEAM-302, BEAM-301 and BEAM-304 edit liver cells inside the body using lipid nanoparticles, for alpha-1 antitrypsin deficiency, glycogen storage disease Ia and phenylketonuria.

The clinical data so far are clean on biology. In BEACON, 31 risto-cel patients reached mean fetal hemoglobin above 60%, per the [NEJM publication](https://beamtherapeutics.gcs-web.com/news-releases/news-release-details/beam-therapeutics-announces-publication-beacon-phase-12-data). At 60 mg, BEAM-302 cut mutant Z-AAT 84% and raised total AAT above the 11 micromolar protective level, according to [ERS data](https://www.globenewswire.com/news-release/2026/09/08/3357432/0/en/beam-therapeutics-presents-updated-clinical-data-from-the-phase-1-2-trial-of-beam-302-in-alpha-1-antitrypsin-deficiency-aatd-at-the-european-respiratory-society-ers-congress-2026.html).

## The odds

Risto-cel is closest to market, with a BLA planned as early as year-end 2026, per the [company](https://www.sec.gov/Archives/edgar/data/0001745999/000119312526331522/beam-ex99_1.htm). The [BIO/Informa/QLS 2011 to 2020 study](https://go.bio.org/rs/490-EHZ-999/images/ClinicalDevelopmentSuccessRates2011_2020.pdf) found that 90.6% of drugs reaching an NDA or BLA were approved, and 93.6% for rare diseases. The risk at this stage is less about approval and more about sales.

BEAM-302 is in a registrational cohort under a planned accelerated approval path. Phase 3 programs have a 52.4% chance of approval in the same study, and rare disease phase 3 programs move to filing 60.4% of the time. A biomarker endpoint that the drug already meets in phase 1/2 should improve those odds, but the plan rests on FDA feedback, not an approval decision. BEAM-304 and BEAM-301 are early. The base rate for a phase 1 program reaching approval is 7.9%, or 17.0% in rare disease.

## The money

Beam held [$1.15 billion](https://www.sec.gov/Archives/edgar/data/0001745999/000119312526331522/beam-ex99_1.htm) in cash and investments at June 30, 2026, and lost $122.7 million in the second quarter. It says cash plus an expected additional $200 million from its Sixth Street facility funds operations into mid-2029.

That facility is [$500 million of secured debt](https://beamtherapeutics.gcs-web.com/news-releases/news-release-details/beam-therapeutics-announces-500-million-strategic-financing) at about 10% interest, maturing in early 2033, with most of it tied to risto-cel milestones. It spares shareholders dilution now, but lenders have first-priority liens on substantially all assets. Revenue is small and lumpy: $0.49 million in the second quarter.

## The risks

Risto-cel enters a market where Casgevy, from Vertex and CRISPR Therapeutics, is already sold and now approved for children as young as 2, per [Vertex](https://investors.vrtx.com/news-releases/news-release-details/vertex-announces-us-fda-approval-expanded-use-casgevyr-treatment). Both need chemotherapy conditioning, which has held back uptake. Beam's BEAM-103 antibody, meant to replace chemotherapy, has only finished a healthy volunteer study.

For BEAM-302, one patient with liver disease had a transient grade 3 liver enzyme rise at 60 mg, according to the [ERS release](https://www.globenewswire.com/news-release/2026/09/08/3357432/0/en/beam-therapeutics-presents-updated-clinical-data-from-the-phase-1-2-trial-of-beam-302-in-alpha-1-antitrypsin-deficiency-aatd-at-the-european-respiratory-society-ers-congress-2026.html). Rival RNA editing drugs, such as Wave's WVE-006, which [lifted Wave's shares 74%](https://pharmaphorum.com/news/wave-surges-first-rna-editing-data-humans) on early data, could compete in the same disease. All three liver programs share one delivery system, so a safety problem in one would raise questions for all.

## Our read

Beam has moved from platform promise to two programs near registration, and the biology in both looks strong. The next 12 months are about execution: filing risto-cel on time, enrolling the BEAM-302 cohort, and turning a sickle cell market that has been slow for everyone into real sales. The [hiring of Eric Foster](https://investor.wedbush.com/wedbush/article/gnwcq-2026-9-1-beam-therapeutics-appoints-eric-foster-as-chief-commercial-officer) as commercial chief shows the company is preparing for that. The balance sheet works, but it is tied to risto-cel milestones and secured debt, so a filing delay would hit both the clinic and the funding plan.
